Objective: To study the effect of Chinese herb Platycodon grandiflorum on roxithromycin pharmacokinetics in blood and drug concentration in lung tissue. Methods: the parmcokinetics and drug concentration in lung tissue were investigated with 312 healthy chickens divided into three groups. Roxithromycin in plasma or in lung tissue was determined after an oral dosage of 8mg/kg bw, 10mg/kg bw and 12mg/kg bw alone or in combination with Platycodon grandiflorum (2g/kgb w),respectively. The concentration of roxithromycin was determined by microbiological assay. The parmcokinetic parameters were caculated by 3P87 PK program; the concentation data in pasma or in lung tissue were stastically analysed by SPSS(12.0) sorftware. Results: The blood concentration-time data roxithromycin in plasma fitted to two compartment model with 1st order absorption. The Cmax values of the experimental groups were significantly higher than those of the control groups, respectively (P<0.05). The Tpeak values of the experimental groups were significantly lower than those of the control groups, respectively (P<0.05). The Ka values of the experimental groups after an oral dosage of 8mg/kg bw, 10mg/kg bw and 12mg/kg bw were significantly higher than those of the control groups, respectively (P<0.05). There were no statistical significant differences of other main pharmacokinetic parameters. Platycodon grandiflorum decreased the time of distribution of roxithromycin to lung tissue, and increased the drug concentration in lung tissue. Conclusions: Chinese herb Platycodon grandiflorum had effects on roxithromycin pharmacokinetical characters and drug concentration in lung tissue.
