In order to investigate the function "lead medicines ascending" of Platycodongrandiflorum we had added the different doses of Campanulaceae decoction andthe isodose tilmicosin for the experimental animals, and Tilmicosin drug concentration inthe lung tissue was detected by high performance liquid chromatography (HPLC), thenanalyzed the drug concentration in the lung tissue. We found Campanulaceae has the roleof "lead medicines ascending".The high performance liquid chromatography (HPLC) was performed in this studyThe Tilmicosin standard chromatographic conditions were established, includingdetermination of the mobile phase ratio of each component, the chromatographic detectionwavelength and velocity. A method of extracting tilmicosin from the lung tissue wasestablished. A standard curve and equation of tilmicosin were estabolished while addedDetermination of Recovery.One hundred and fifty one-day-old chickens were randomly divided into A, B, C,D, Egroups, each group had30chickens. After one week, Experimental groups wereadded Platycodon grandiflorum solution that was diluted at the concentrations of3g/kg(A,high dose),2g/kg(B, medium dose),1g/kg(C, low dose), D was control group, and E wasblank group. At day28, chickens of A, B, C and D groups were injected withtilmicosin injection, and birds in group E were not injected. Then we quicklysacrificed the chickens at the time points of1.0h,1.5h,2.0h,2.5h,3.0h,4.0h,6.0hrespectively, lung tissues were sampled after washing with normal saline, extractedTilmicosin from lung tissue, and took it for HPLC analysis. The results show that theadding platycodon grandiflorum groups (A,B,C) had higher drug concentration at all timepoint than without platycodon grandiflorum group(D), and B group had the highest drugconcentration than other groups.Conclusion: Campanulaceae can affect pharmacokinetic parameters of tilmicosinand the drug concentration of lung. And Campanulaceae can improve tilmicosinconcentration of the drug in the lungs, the most obvious effect is middle dose, followed ishigh dose, and last is low dose, so we can verify patycodon grandiflorum "drugloading upward" effect.
