Study On Technique System Of The Tissue Culture In Bougainvillea Glabra

The stem segments of Bougainvillea glabra were used as explants in study of organ culture to establish a rapid technique system of propagation in vitro and to find out the best explants,medium culture procedure and the possible ways of plant regeneration .The results were as follows:1) The best explants were stem segments which can be obtained from a wider area and was easier to be sterilized.April was the best period of picking the stem segments in this test.2) The best initiation medium was MS+BA1.0 mg/l+IBA0.5 mg/1.After 30 days culture ,the inducement rate could be up to 95%;and the average number of axillary buds could reach 6.0.3) The multiplication of callus induced shoot clumps: MS+BA2.5 mg/l+IBA0.2 mg/l+GA31.0 mg/l was the best medium.The multiplication rate of shoot clumps could be up to 7.7.4) The multiplication of side buds: MS+BA3.0mg/ l+IBA0.1mg/ 1 was the best medium,the multiplication rate of side buds coud be up to 4.0.5) As a result of the comparing of the two shoot clumps multiplication,the way of side buds was superior.6) The best medium for rooting of shoots was 1/4MS+IBA1.5 mg/ 1+NAA0.05 mg/ 1+AC0.5 mg/1 ,in which the root of rooty plantlets grew rapidly and the rooting rate could reach 95%.7) The better rooting agent was ABT and 50 mg/l was the suitable concentration,withwhich the rooting rate could be up to 48.3%.8) For nursling of plantlets.the medium witn 1/4 ruby mica and 3/4 soil of the rottenleaves in the bamboo groves was the best materials,the survivial rate of plantlets could beup to 90%.
