Sarcandra glabra was exployed as the experimental material to carry on the in vitro propagation and establishment of regeneration system from leaves. Through the serial treatments about the comparison of different explants,the effect of medium,carbon source,GA3,plant growth regulators on the growing of shoot were explored in tissue culture.So the in vitro propageation system was established.1.The culture of stem apex inductionUsing mewly developed shoots with byds as a source of explants could facilitate the establishment of the asepsis system.The best way of sterilization was putting shoots in 0.1%HgCl2(adding several drops of Tween 80)for 10 mins.It has great effect of the elements of medium on the establishment of in vitro system.MS was chosen as basic medium in the following experiment,in which the ability of differentiation was stronger,and the buds grow better.The result showed that sucrose was the best carbon source of medium,and the best concentration of sucrose was 20g/L.Sprouts could be induced in MS mediums surplus with 6-BA,KT,NAA,IBA,among which MS+6-BA2.5mg/L was the best.2.The culture of bud proliferationThe cluster byds were cut into a single bud and cultivated in the sub-enrichment culture medium MS adding different concentration of 6-BA and NAA.The best medium is MS supplemented with concentration of 6-BA2.0mg/L,NAA0.3mg/L and GA31.0mg/L.The rate of shoot proliferation was6.14,and the bud grow healthy and strong.3.The culture of rooting1/2MS medium supplemented with proper concentration of NAA or IBA can induce rooting.1/2MS+IBA1.0mg/L+NAA0.2mg/L medium are suitable for indecing roots.4.Plantlets transplantationThe survival rate was affect by transplantation invirenment.The survival rate cultivated in sand of rooting plantlets could be up to 70%.The transplantation environment played an important role for survival rate of tube culture-plants,the result showed that the survival rate was higher relatively for Sarcandra glabra culture-plants,transplantated under forest which had the similarly environment with former plant,and survival rate could reached to 70%transplantated in river sand.
