Bougainvillea,abundant of varieties are popular in landscaping with strong adaptability,easy cultivation,beautiful flowers and leaves,diverse color,long flowering and high ornamental value.In addition,lots of chemical substances discovered in Bougainvillea are helpful on human body,life environment,which are important medicinal raw materials for antiviral.Currently,Bougainvillea is considered as city flower by 18 cities of Beihai,Wuzhou,and Shenzhenso no.With the progress of society,market demands are increasing on Bougainvillea,as an important landscape plant,due to the people demanding on the urban ecological environment.t.Nowadays,asexual reproduction,such as cuttings and layering,on Bougainvillea had the disadvantages of long production cycle and unable to meet the needs of the market.Based on this,study on explant disinfection,primary culture,proliferation culture,strong seedling culture,rooting culture,and transplanting were carried out in 3 varieties of Bougainvillea,Bougainvillea tissue culture system was initially established,which would provide technical support for the large-scale production of Bougainvillea.The results showed that:1?Bougainvillea explants can be contaminated with mold and bacteria,mold contamination was introduced in the operation process,whilst,bacterial contamination came from its own endophyte.A suitable method for 3 varieties was found out in 36 disinfection combinations:75%alcohol disinfect in 30-35s,0.1%mercuric chloride disinfect 6-8min,and 0.1%Twain.2?In primary culture,callus was induced from 3 varieties of Bougainvillea's leaves,stems and buds on the 12 starting medium,in which,shoots could be induced by stems and buds.The induction rate trends of 3 varieties in 3 kinds of explants were Bougainvillea spectabilis 'Crimsoniake'>Bougainvillea glabra'Rosa'>Bougainvillea x buttiana 'Barbara Karst'.The results showed that MS medium was more suitable for inducing callus and shoots of leaves;the concentration of 6-BA and NAA which was suitable for inducing Bougainvillea callus or bud was 2.0-2.5mg/L and 0.05-0.2mg/L respectively,and NAA was better than IAA.3?It was difficult to induce buds and lateral buds by the way of callus,but they could be induced through the pathway of buds,the proliferation coefficient of Bougainvillea 'Crimsonlake' was higher than Bougainvillea glabra 'Rosa'and Bougainvillea x buttiana 'Barbara Karst',Bougainvillea x buttiana 'Barbara Karst' was the lowest.When the concentration range of 6-BA and NAA was 2.0-2.5mg/L and 0.05-0.1mg/L,Clustered Shoots could be induced,and the proliferation coefficient was higher when used NAA than that of IB A.4?The medium MS+6-BA1.0mg/L+NAA0.2mg/L was suitable for seedling medium,and the growth rate of the seedlings on this medium was higher than that of other medium,and the seedlings were more vigorous.5?The highest rooting rate of Bougainvillea glabra 'Rosa'?Bougainvillea spectabilis 'Crimsonlake'and Bougainvillea × buttiana 'Barbara Karst'were 39%,46%and 23%,the corresponding medium was 1/2MS+IBA0.5mg/L+GA 31.5mg/L?and?1/2MS+IBA0.3mg/L+GA31.0mg/L?1/2MS+IBA1.0mg/L+GA 32.0mg/Lrespectively.
