Zmglpl gene is cloned from maize, which expresses especially in the young green leaves. This paper focuses on the expression analysis of the gene, and the deletion mutution of the promoter. It is transformed into Arabidopsis thalianawith the reporter gene, GUS, and a lot of satisfying results have been concluded.It has revealed that different deletion fragments have different circadian rhythm, and each shows its especial expression activity in different organs. The activities of A 637, A 1027 and A 1215 become stronger gradually when exposed to the sunlight in 8 hours, and weakergradually in the darkness. The fragment?0nt-98nt includes theessential element to regulate the gene expression in green organs andflower silk, but 171nt-70nt in A 1215 sequence has similar elementin the root. It seems the fragments -70-171nt and -359-749ntinclude the sense regulator to enhance the expression activity, but theuppersteam fragments 171-359nt and 749-1255nt restrain that.Or there are sense regulators in the fragments 70nt-98nt and 171359nt, but their upperstream sequence 70-171nt and 359-749nt can restrain the anti-sence regulation.A 637 sequence has a higher activity than others. When exposed to 16 h light/8 h darkness, its average activity is three times as high as the known 35S CaMV.These results will promote further researches such as the drive of the expression of insect - resistant genes in the plant and vegetablebiotechnology, especial expression analysis in different organs and circadian rhythm of these regulators.
