Wet damage was caused by higher water content in soil.It seriously affected growth of barley and induced to fall off to some extent in yield all over the world,especially in the middle and lower reaches of the Changjiang River in our country. Researches about waterlogging-tolerance heredity and molecular markers in barley are not enough.So it is necessary to do research about waterlogging-tolerance for meeting the requirement in breeding plant new waterlogging-tolerant barley. In this study , F2 population(StirlingXJingluo 11)was used to develop primarily SSR marker that was linked towaterlogging-tolerance in barley.At present, waterlogging-tolerance index include plant height,spike number,weight of 1,000 grains, physiological indexes include cholophyll content etal . However, there are not uniform indexes.This research was studied based on morphological and physiological characters for screening suitable waterlogging-tolerance indexes in barley. We used 3 F2 populations to carry on with experiment(a population was treated by water,other two as check). We analysised frequency distribution of plant height.spike number,weight of 1,000 grains and cholophyll content after treating by water in 3 F2 populations .Results showed frequency distribution of the cholophyll content on second top of leaf with normal distribution before water treatment .After water treatment, thedistribution of cholophyll content changed into two normal distributions.This result can reflect on the difference for waterlogging-tolerance in each plant. It could be a suitable index for waterlogging-tolerance in barley.Using 73 SSR primers ,we choosed Stiring and Jingluoll as the parents among 13 waterlogging-tolerant breed and 13 waterlogging-sensitive breed in barley.Many scholars indicated that waterlogging-tolerance was dominated by polygene in barley. So it was very difficult to exactly map these genes. It was a good method to map waterlogging-tolerance through BSA (bulked segregant analysis).According to result of measurement of cholophyll content, we choosed 7 extreme waterlogging-tolerant individuals and 7 extreme waterlogging-sensitive individuals to construct respectively tolerant pool and sentive pool . The result of PCR amplitified showed the polymorphism between the pools and the parent was coincident .Use software of MATLAB V6.5 to analysis ,we found the marker WMC1E8 can explain the variation 29.91% for waterlogging-tolerance in barley. According to the Barley Genomics Unit issued by the Scottish Crop Research Institute ,we found the marker WMC1E8 was located in chromosome 1 in barley , the result showed waterlogging-tolerance main effect gene probably lies in chromosome 1 in barley.
