The material of the following studies is colored calla lily (Zantedeschia hybrida), belongs to the family Araceae and the genus Zantedeschia. Key techniques of rapid propagation in vitro and treatments of cultivation were investigated. 1. In this study, Zantedeschia hybrida 'Parfait' were used as experiment material. In nature, Zantedeschia grows in marshy habitats that provide a good climate for the growth of rotting bacteria and fungi. So explants were pretreated with different fungicides or hot-water to reduce the contamination. Then they were inoculated into the medium with different antibiotics, in order to find a good treatment to remove pollutants from explants in the initial culture. Next experiment was to induce adventitious bud with the orthogonal design, and to find what influence on proliferation of adventitious bud under the treatments of different types and concentrations of cytokinin. After that, different exogenous phytohormones were investigated to see which treatment was good for adventitious bud to rooting and growing. Then we should transplant these plantlets into different substrate with different strength of sun shine. We also studied how to induce microtuber with different plant regulator and cultural condition. 2. The influences of different cultivation treatments on height growth of 8 cultivars of colored calla lily were investigated. On the other hand, we used 200~400mg/L PP333 and 1000-2000mg/L CCC on two cultivars of potted colored calla lily to see if the plant would be dwarfed.The results showed:(1) decrease the pollute rate in initial tissue cultureUsing 3g/L Captain fungicides and pretreating explants 24h, we found that the pollute rate was very low. And it is better than the treatment of hot-water. The medium which added 100mg/L Penicillin+100mg/L Gentamycin, had good effect on decreasing the pollute rate.(2) induce and proliferation adventitious budsThe best treatment of induce adventitious bud was 20g/L sugar + pH 5.8 + B5 medium. After added cytokinin into the medium, the induced rate jumped to 100%. Whenadded BA 3mg/L, the proliferation effect is the best.(3) growing, rooting and transplanting of plantletNAA 0.5mg/L+IBA0.1mg/L+MS medium was the most useful treatment to plantlet's growing. The rate of fresh weight growth was the maximum. The most useful treatment to plantlet's rooting was NAA 0.1mg/L+IBA0.5mg/L+MS medium. After transplanting, the survival rate of plantlets which were planted under 1 layer shading net was great.(4) the factors of microtuber inducingWhen there was 50g/L sugar in the medium and 16h/d light, the induction of microtuber was the best. When added plant regulator into this medium, we found: added BA, the induction rate was up to 100%. Especially added 3mg/L BA, its induction effect was the best.(5) different treatments of cultivation on height growthThe difference of cultivate-substrate didn't affect much on height growth of most cultivars. On this basis, 2 cultivars, 'Crystal Blush' and 'Parfait', were used in the experiments of different concentrations of fertilizer and different rates of N, P and K. The results showed that there is no significant difference among each treatment of different concentrations of fertilizer, especially among those in later period of growth. The treatments of different rates of N, P and K showed that the change of N content has little effect on height growth; but increasing of P and K contents promoted height growth of colored calla lily.(6) dwarfing experiment with PP333 and CCCUsing 200~400mg/L PP333 and 1000-2000mg/L CCC on two cultivars of potted colored calla lily, the heights of them were obviously dwarfed after one month sprayed weekly, and the plant stems grew thickly. The results of statistic analysis show that PP333 is more effective than CCC in dwarf culture, and the optimum concentration of PP333 is 300mg/L. It suggests that PP333 has applied value in dwarfing treatments of colored calla lily under facilities-cultivation.Colored calla lily reproduces through tubers which results in low reproductive efficiency, diff...
