Zantedeschia hybrida belongs to the araceae zantedeschia bulb flowers,it was a relative of calla lily,its leaf,color all beautiful,it was important potted flower and cut flower material,extremely popular in the domestic and foreign markets,has the very big development space.The traditional breeding method of the Zantedeschia hybrida,basically have seed to breed and divide a ball to breed two kinds,due to long breeding period,propagation coefficient was low,plant growth condition was poor,susceptible to bacterial soft rot and spreading the virus,it was difficult to meet the production needs.In search of a solution,in this study,'Murano','Memphis','Kelso','Blade' and 'Salada' were used as the experimental materials,the effects of different pretreatment methods on the reduction of contamination rate in primary culture were studied,and the effects of different plant growth regulators on the rapid propagation in vitro,in order to establish a complete rapid propagation system in vitro,finally,five varieties of tissue culture seedlings were used to study the culture of inner ball,to compare the effects of different factors on the nodules in the bottle,look for key impact factors,it provides a theoretical basis for the induction of miniature seed balls in the bottle of Zantedeschia hybrida,the main research results are as follows:1.Screening of pretreatment methods for explants of different varieties of Zantedeschia hybridaThe suitable sterilization time of 'Memphis' and 'Blade' was 20min,and the best sterilization time of 'Murano','Kelso' and 'Salada' was 18min.Through a filter for different pretreatment methods,appropriate'Memphis','Salada' of pretreatment method was 1.5 hours,45? hot water bath treatment was suitable for 'Murano','Kelso','Blade'pretreatment method was bactericide 500 times liquid handling 4 hours.2.In vitro rapid propagation of different varieties of Zantedeschia hybrida(1)Zantedeschia hybrida induction in callus of calla hybrida:the best callus induction medium for 'Murano' and 'Kelso' was MS+6-BA1.0mg/L+NAA0.2m/L,and the induction rates were 56%and 47.67%,respectively.The optimal callus induction medium for 'Memphis' and 'Salada' was MS+6-BA 1.0mg/L+NAA0.3mg/L,and the induction rates were 53%and 54.67%,respectively.The optimal callus induction medium for blade was MS+6-BA2.0 mg/L+NAA0.2 mg/L,with an induction rate of 78%.(2)Adventitious bud induction of calla hybrida:MS+6-BA3.0mg/L+NAA0.3mg/L+KT0.3mg/Lwas the best adventitious bud induction hormone combination of 'Murano' and 'Blade'MS+6-BA5.0mg/L+NAA0.5mg/L+KT0.2mg/L was the best combination of adventitious bud induction hormone in Memphis.MS+6-BA5.0mg/L+NAA0.1mg/L+KT0.3mg/L was the best combination of'Kelso' adventitious bud induction hormone.MS+6-BA5.0mg/L+NAA0.3mg/L+KT0.1mg/L was the best adventitious bud induction hormone combination of 'Salada'.It was found that 6-BA had the greatest influence on the induction of adventitious buds.With the increase of 6-BA concentration,adventitious buds gradually increased in height,and the growth state was good and robust.(3)Buds proliferation and culture of calla hybrida:The results showed that the hormone concentration was too high or too low.A combination of high concentrations of the hormone promotes bud proliferation but inhibits seedling growth.The optimal combination of 'urano' in the proliferation culture of the buds was MS+6-BA3.0mg/L+TDZ0.05mg/L+KT0.3mg/L.The optimal combination of'Memphis' and 'Salada' was MS+6-BA3.0mg/L+TDZ0.2mg/L+KT0.2mg/L.The optimal combination of 'Kelso' and 'Blade' was MS+6-BA2.0mg/L+TDZ 0.1mg/L+KT 0.3mg/L.(4)Rooting induction of colorful calla lily:the optimal rooting induction medium for 'Murano','Memphis' and 'Blade' was 1/2MS+NAA0.5mg/L+IBA0.3mg/L;The optimum rooting induction culture for 'Kelso' and 'Salada' was 1/2MS+NAA0.5mg/L+IBA0.5mg/L.3.Research on the technology of inner balling of Zantedeschia hybrida(1)Different sugar concentration and multi-effect azole combination of Zantedeschia hybrida knot ball bottle impact studyThe combination of high concentration of sucrose and low concentration of pleurozole is h elpful for the formation of small pellets in the bottle.Suitable for 'Murano','Memphis','Blade'ball formation within the optimal hormone combinations was MS+NAA 0.5mg/L+Sugar 80g/L+Multi-effect azole2mg/L;Appropriate 'Kelso','Salad' of pellet form of the optimal hormone com binations in the bottle was MS+NAA0.5mg/L+Sugar 80g/L+Multi-effect azole4mg/L.(2)Different culture medium type and concentration of NAA combination of Zantedeschia hybrida knot ball bottle impact studyThere were significant differences in the nodulation rates of different cultivars in different media.The optimal medium combination formed by pellets in 'Murano','Memphis','Blade',and'Salada' bottles was MS+Peat120g/L+Perlite40g/L+NAA0.8mg/L;Appropriate 'Kelso' ball bottle forming combination is the best medium MS+Agar 6g/L+Sugar 80g/L+NAA0.5mg/L.(3)Combination of different light intensity and illumination time research on the influence of Zantedeschia hybrida knot ball bottleThe light combination suitable for the formation of small balls in the bottle of 'Murano','Blade'and 'Salada' was 20001x light intensity and 16h light time,the light combination suitable for the formation of small balls in the 'Memphis' bottle was 40001x light intensity and 8h light time,the light combination suitable for the formation of small balls in 'Kelso' bottle was 40001x light intensity and 12h light time.
