Rice is one of the most important crops, which provides main food for more than one half of the world population. Brown planthopper(Nilaparvata lugens Stal, BPH) is one of the most serious pests of rice in Asian . The use and breeding of resistant breed to BPH is an effective and economical way of controlling BPH. B3F4 is a near isogenic lines(NIL) screened from Cv 1665 of oryza officinals ♂ X Cv Gui99 of oryza sativa In this study, B3F4 was used to study the molecular marker of Bph derived from Cv 1665.The main results are as follows:l.According to results of resistant identification of B3F4 plants, we constructed the DNA pools of resistance and nonresistance. 282 arbitrary primers were used to screen RAPD marker linked to Bph.We have obstained 5 RAPD markers linked to Bph: BS294, BS229, BS303 BS403 and BS1159, and 5 RAPD markers were used to finish analysis of RAPD markers of B3F4 plants;2.With cloning and sequencing of 5 RAPD markers, we obstained nucleotide sequence and length of RAPD markers: BS294 821bp BS229 703bp, BS303 649bp BS403 783bp and BS1159 1408bp;3. According to nucleotide sequence of BS229 and BS1159 , we designed two pairs specific primers to complete SCAR marker analysis transformed from RAPD marker. The results show SCAR marker analysis is in accordance with RAPD marker's.4. According to The results of RAPD marker analysis of B3F4 plants, with bioinformation soft MAPMAKER/EXP.Version 3.0,we constniced regional link map among 5 RAPD markers and Bph, it covers 18.2cM region. The map is useful for precise lacalization and cloning of Bph.
