By means of seedling selection ,the author bred and selected 14 ground-cover chrysanthemum new cvs with different blooming times.In order to get Haploid plants of chrysanthemum for breeding needs of this study, Chrysanthemum cultivars 'A9-14' was used for anther culture. The effects of genotype, pretreatment, sucrose concentration, types of media and the addition of various growth hormones were analyzed. Also, the cytology of plants regenerated via anther culture was observed. Results :1 .Relationships between microspore stages and morphology of florets were observed. When florets color is white, anthers contained more microspores at the uninucleate stage.2.The effects of genotypes on the anther culture are obvious. In this study, anther cultures of five genotypes 'B12', 'C18', 'D14', 'E1', 'A9-14' is observed and genotype 'A9-14'has the highest induction ratio at 21.4%.3.The most effective medium for genotype 'A9-14' is MS+BA 2.0mg/L+2,4-D 1.0mg/L, plus with 9%W/Vconcentration of sucrose.4.Callus differentiation is best on the medium MS+BA1.0mg/L+ NAA0.1 mg/L +2%W/Vsucrose.5.A pretreatment at 4℃ for 48h enhances the induction ratio. The addition of activated charcoal (AC) to medium greatly reduces callus formation.6. Pollen plants used in the study are mostly hexaploid (2n=6x=54), minorities are triploid (n=3x=27).7. The callus formation and regenerated plants of cultivar 'A9-14' are obtained for the first time when isolated microspores are cultured in liquid medium.
