| The sacculus rotundus having various functions such as immunity, endocrine and assimilation, are only found in rabbits. In order to elucidate development of the diffuse neuroendocrine (DNES) and the related immune cell in sacculus rotundus of rabbits, the studies, using the methods of immuno-histochemistry, electron microscopic cytochemistry and so on, observed the distribution and quantity of substance P (SP), synaptophysin (SYP), protein S-100, CD3 (T lymphocyte), CD20(B lymphocyte), CD57 (NK lymphocyte) and CD68 (macrophage) in the sacculus rotundus of 1-60th day's rabbits. Moreover, the sacculus rotundus are observed by means of SEM in 5th and 15th days' rabbits. Here follows the results.The progressive increasement of positive cells with the substance P (SP), synaptophysin (SYP) and protein S-100 was observed in the lymphoid tissues of sacculus rotundus of rabbit in this study. But there was variousness of emergence time and place in the sacculus rotundus. Meanwhile, it can be ensured that DNES cells are related to maturity of the lymphoid cells.Based on the histochemistry observation, there appears two types of cell in the lymphoid tissues of sacculus rotundus in the 1st day: A type (named type I cell) showing large, pale, round or oval shapes, has large and bright nuclear, but little cytoplasm. A great number of slender processes were observed, and the protrusions between the cells interweave each other, forming loose reticulated formatio. There are more the cells in the early development of sacculus rotundus; The other type of cells (named type â…¡ cell) are mostly of dark color and small size. The number of type â…¡ cells are less than type I in the early development of sacculus rotundus. At the 5th day, there appear some round lymphoblasts with large, dark nucleus in the lymphoid tissues of sacculus rotundus. The lymphocytes and macrophages increase progressively.Through observation and quantitative analysis for different days' sacculus rotundus of the T lymphocyte (CD3), B lymphocyte (CD20), NK lymphocyte (CD57)and macrophage (CD68), it was found that, at the 1st day, there appear a number of CD68~+ and CD57~+ cells other than CD3~+ and CD20~+ cells; at the 10th day, there appear few CD3~+ and CD20~+ cells; as rabbit growing, CD68~+, CD57, CD3~+ and CD20~+ cells are all increased obviously; at the 40th day, CD3~+ cells and CD20~+ cells are more than CD68~+ cells and CD57~+ cells notably (P< 0.05). It is showed that there are certain correlations between these types of cells by SPSS.Using SEM method, it was found that the sacculus rotundus of the 5th day's rabbits have smooth surface, sparse villuses, in the top of which there are a number of regular micro-foramina. In the 15th day, the villuses present a shape of finger and have rough surface in which there are less foramina. These morphologic transform may be related to the evolution mechanism of nourishment and defense in infant rabbits.The analytical results above showed that the three indicators of DNES cells appear in different time and place of the sacculus rotundus. The evidence of related immune cells implied that emergence time of acquired immunity cells is later than natural immunity cells in the lymphoid tissues of sacculusrotundus, and that the sacculus rotundus are peripheral immune organ.The research mainly showed growth characteristic of DNES cell and immunity-related cell in sacculus rotundus of infant rabbits, supplied a new morphologic evidence for further study to Immune-neuro-endocirin network (I-N-EN ) theory of mammal animals and for controlling of rabbit diseases, especially infectious intestine disease of infant rabbit.
