In this study, genetic diversity in 19 lines of yellow-seeded rape (Brassica napus L.) has been studied using isozyme, Esterase and Peroxidase, and SSR markers. In addition, the fingerprinting of 5 varieties in .yellow-seeded rape (Brassica napus L.) has been constructed. The detail results were as follows.1. 19 lines of yellow-seeded rape (Brassica napus L.) were assayed by two isozymes, esterase(EST), peroxidase(POD), with Polyacrylamide Gel Electrophoresis. Five allzymic loci and 15 alleles were detected. All the allzymic loci were polymorphic. The frequency distribution of allzymic loci in rapeseed range from 0.3158-1.000, and the average heterozygosity of five allzymic loci was 0.4627. The result of cluster analysis UPGMA with the isozyme data suggested that 19 lines could be divided into four groups, the genetic distance between four groups was about 0.38. The study showed that yellow-seeded rape (Brassica napus L.) had high genetic diversity in terms of isozyme.2. Genetic diversity in yellow-seeded rapeseed was investigated using forty-two SSR primer pairs. A total of 336 polymorphic and clearly scorable bands were generated. The number of bands per primer pair ranged of 2 to 21, with the average of eight. The value of polymorphism information content(PIC) varied from 0.17~0.85,with an average of 0.73. The genetic similarity cofficient of 19 lines varied from 0.63~0.87, with average of 0.72. Through cluster analysis based on the 42 SSR markers by UPGAM, the 19 lines could be classified into four groups, which is generally agree with the pedigree analysis.3. The fingerprinting of 5 varieties in yellow seeded rapeseed (B. napus L.) of China was constructed by SSR marker technique in order to distinguish truth or falseness of seeds.
