A Study On Tissue Culture Of Osmanthus Fragrans Lour

The purpose of the study was to primarily establish the system of effective tissue culture of Osmanthus fragrans Lour. The study took the buds in a dormant state in winter﹑germinating buds﹑the shoots and stems of young branchs and embryoes at the different stage of growth as the explant, puts more emphasis on the most suitable time of getting explants, the most favotite fostering condition as well as the effective measures taken against the browning and contamination. The result is shown as follows: (1) The best explants are the germinating or the top buds of newly protruding tender branch while the top buds mustn't stop growing. (2) In initial culture ,it can effectively prevent the explant from browning to append 0.5g/L PVP to culture medium. (3) The initial culture: the more favorite initial culture medium of germinating buds is B₅+6-BA₅.0mg/L+NAA0⁰.05 mg/L+30g/Lsuorose, the rate of initiation can reach 84.4%; the best medium of top buds of little shoots stems is B₅+6-BA3.0mg/L+NAA0⁰.05 mg/L+30g/Lsuorose, the rate of initiation could reach 80.0%. There exists different culture condition in different genotypes. (4) The multiplication of budds: We found in experiment the culture materials didn't generate cespitose buds. Even through high dense of 6-BA was offered. The most suitable multiplicated medium is B₅+6-BA₂.0mg/L+NAA0⁰.05 mg/L+30g/Lsuorose. (5) The medium suitable to root for plants introduced budds or top shoots is B₅ +NAA2.0 mg/L30g/Lsuorose. (6) After the fruits are ripe,the seeds need long time dormantation the tissue culture can enhance the budding of the seeds in advance. The suitable time of getting explants is when the endosperm has ossified to the complete ripeness of the fruits. (7) The medium suitable for the induction of embryos is : B₅+6-BA₂.0mg/L+NAA0⁰.05mg/L+30⁵0g/Lsuorose. The lightness is superior to the darkness. (8) The suitable culture medium to elongate is B₅+6-BA1.0².0mg/L +30/Lsuorose. (9) The suitable rooting medium of plants introduced from embryos is B₅+IBA₂.0mg/L+30/Lsuorose.
