The plant expression vectors of the FMDV structure protein gene P1 have been transformed into potato by our laboratory. The positive result was obtained after PCR and PCR-Southern analysis. On account of these results, the main factors influencing genetic transformation into potato were optimized. The P1 gene was transformed into potato different varieties and explants through different methods. The target genes expressed in the transgenic potato plants. This will provide theories or methods for producing new gene engineering subunit vaccines using transgenic plant as bioreactors. Different potato explants of Favorita and Dongnong 303 were transformed by Agrobacterium tumefaciens with gene FMDV P1. 174 resistant putative plantlets were obtained after kanacyllin selection, which included 91 resistant plantlets of Favorita and 83 of Dongnong 303. 17 positive plantlets were obtained by PCR and PCR-Southern. The results showed that the transgenic potato plants with P1 gene were obtained. Different potato explants of Favorita and Dongnong 303 were transformed by gene-gun with gene P1. 116 resistant putative plantlets were obtained after resistant selection. 10 positive potato plants were obtained by PCR and dot Blot techniques. All the transgenic positive potato plants with gene P1 were tested by SDS-PAGE. It was found that unique band of 31 kD existed with SDS-PAGE in 7 transgenic plants.
