Male And Femal Origin Of Four Chinese Yellow Cattle

In order to investigate the male and female origin of 4 yellow cattle,10 microsatellite loci of Y chromosome, Cytb gene of mitochondria DNA and HMG-box of SRY gene were analyzed by microsatellite DNA marker techniques, PCR-RFLP and PCR-SSCP techniques. The results were as follows: 1 10 microsatellite loci were studied in this research. We found that there were 2 alleles in INRA124 locus. In UMN2404 locus, yellow cattle had 3 alleles: 85 bp,63 bp and 55 bp, and yak had 55 bp allele, 55 bp allele had a low frequency in yellow cattle. There were no polymorphisms among yellow cattle and yak respectively, but yellow cattle and yak had different allele, so this locus could be used to differentiate yellow cattle and yak. In BMS861locus,UMN0504 locus,UMN1203 locus and UMN3008 locus, there were no polymorphisms among yellow cattle and yak, and yellow cattle and yak had the same allele. In UMN2303 and UMN0920, we only got smear band. 2 After microsatellite detecting INRA124 locus, we found that Nanyang cattle,Luxi cattle,Jinnan cattle and Qinchuan cattle all had Bos Taurus and Bos Indicus male origins. Nanyang cattle had 72.73% Bos Indicus male origin and 27.27% Bos Taurus male origin. Jinnan cattle had 23.33% Bos Indicus male origin and 76.77% Bos Taurus male origin. Luxi cattle had 75% Bos Indicus male origin and 25% Bos Taurus male origin. Qinchuan cattle had 25.8% Bos Indicus male origin and 74.2% Bos Taurus male origin. Nanyang cattle and Luxi cattle had high frequency of Bos Indicus male origins, and Qinchuan cattle and Jinnan cattle had Bos Taurus male origin. 3 There were 3 alleles in UMN2404 locus, but we found that these 3 alleles all had no special frequency among these 4 populations, they also had no special frequency between Bos Taurus and Bos Indicus male origins cattles. 4 We had 6 heliotypes from INRA124 locus and UMN2404 locus. H1 heliotype had a high frequency in Nanyang cattle and Luxi cattle, and H2 heliotype had a high frequency in Jinnan cattle and Qinchuan cattle. H6 heliotype did not found in Nanyang cattle and Luxi cattle. 5 Yellow cattle and yak had different allele in UMN0307 locus, so this locus could be used to differentiate yellow cattle and yak. We did not find yak's allele in yellow cattle populations, which indicated that there was not yak male origin in these 4 populations. 6 After PCR-RFLP analyzing 271 bp sequence of mitochondrial DNA Cytb gene, we found that yellow cattle all had HinfI digesting locus, but yak hadn't. So this indicated that this sequence cloud be used to differentiate yellow cattle and yak, and it also indicated that there was no yak female origin among these 4 populations. 7 After PCR-SSCP detecting partial sequence of SRYgene HMG-box, we found that there were no polymorphisms among yellow cattle and yak, there were even no polymorphisms between yellow cattle and yak. And this indicated that this sequence was highly conserved.
