RIL (recombined inbred line) populations were very useful in plant breeding and genetic research. Furthermore, as a kind of permanent population, the RIL population could be evaluated in many different environments. So the genetic component of variance and the interaction between QTL and environment could be estimated more accurately when the quantitative traits were studied.Jindou23 as a mother, Huibuzhi as a father in this research .A cross of Jindou23 x Huibuzhi is made, then selfed for 13 generation by single seed descent method to construct RIL populations. SSR markers are used in evaluating the population. Evaluation shows that almost all of SSR loci and lines are purified. The distribution of genotypic composition of the population fits normal well. The population is suit to genetic mapping and otter genetic research for its well-dispersed genetic structure.The study aims at choosing SSR makers with the connection of oil.At the same time , correlation traits and markers with oil are located .333 pairs of SSR primers in the study, which cover almost the whole soybean genome, are used to select Satt259, Satt353, Satt119, Satt533, Satt 05, Satt 491, Satt 199, Satt038, in addition eight pairs of pairs different primers by BSA pool. With the help of Doctor Yongjun Wang in Institute of Genetics and Developmental Biology Chinese Academy of sciences, by means of soft analysis of soybean seed oil trait, 3 QTL are detected and located in Genetic map drawed by him.Satt491 with connection of oil is located E link group,267.9 cM location, taking on additive effect .Satt491 can be used in the breeding of high oil soybean by molecular marker, gene clone and transplant gene study. Sattll9 is located in A2 link group. Satt199 is located in O2 link group.
