| With RAPD and ISSR method,the paper studied genetic diversity and differentiation of two natural population of Pinus koraiensis in Liangshui National Nature Reserve and Fenglin National Nature Reserve. Through the amplification of RAPD with 10 random primers, 61 repeatable loci were detected, and there were 103loci in amplification of ISSR with 13 primers, and ISSR could examine more heredity polymorph than RAPD.(in the RAPD amplification and percentage of polymophic loci was 0.6935%. Nei's index showed the levels of genetic diversity of Liangshui population(0.2634) was higher than Fenglin'(0.2607); in the ISSR amplification and percentage of polymophic loci was 79.61%.genetic variation existed mainly within populations, there were little genetic differentiation between populations. The level of genetic diversity that the Korean pine is in the loose department plant is not low that studies have shown. Therefore explain why the species of the Korean pine is endangered , the distributing area narrows gradually, it is because of human destruction function after all , in addition, such factors as the fire and wind pour cause. |
