Korean pine (Pinus koraiensis) can provide us with not only valuable timber, but also famous, high-quality and special dry fruit. Its pine nut contains a large amount of biological active composition and rich nutrition. DNA was extracted from the leaves of Korean pine by improved CTAB method. The purity of DNA that extracted by this method was high. he best stable reduplicative ISSR-PCR reaction systems of Koreanpine and PCR amplification parameters were established fourteen ISSR-PCR primers that had vivid amplification band and abundant polymorphism had been screened.Forteen primers were screened out with ISSR technique, and 90 sites were obtained totally from three populations of Pinus koraiensis, an introduction as material, in different geographical distribution zone.72.22% of which, namely 65 sites, belonged to polymorphic sites. Result of genetic diversity suggested that the ratio of polymorphic sites of populations was 72.22% in average, and the genetic diversity was abundant judging from the average value of Shannon index 0. 4461. Gene flowing and genetic differentiation existed among populations considering from the value of Nm was 4.5062, the average value of Nei index 0.3005 and Gst index 0. 0999. Genetic diversity among populations possessed 90.01% of the total and genetic diversity between populations possessed9.99%.we can conclude that Genetic diversity mostly esit inpopulations.besides, there were some relations between genetic distance and geographical distance.All results of this study will provide the theoretical basis for the selecting of improved Korean pine variety and evaluatione of genetic diversity in the artificial forest...
