Factory Production Technique For Culture Of Populus Tremla×P.Tremuloides

In this paper, elite plants of Populus tremula ×P.tremuloides were used as research material. A series of experiments were carried out in order to set up a micropropagation technique of the hybrid aspen for commercial purpose. Such as stablish of tissue culture system optimization, simplifying of the culture in vitro rooting and cuttage, to make the cost of produce lower. The conclusions were as follows:1. Rapid propagation by multiplication of axillary buds: The optimum culture medium of differentiation was NT+BA0.1mg/L+KT0.5mg/L+2%sucrose, the rate differentiation was 100%, a single explant has 6.9 buds averagely; WPM+NAA0.3mg/L+IAA0.05mg/L+2% sucrose was the optimum culture medium of rooting. The rooting rate was more than 93%; the suitable medium of test-tube transplants was the mixture of humus and sand, which is mixed according to the scale of 2:1. The medium was sterilized by high temperature or agricultural chemical. The optimum air temperature for transplanting is 18—26℃,The moisture in the air should be kept above 60% by leaf spraying during the first 5 days after transplanting. In the experiment, the survival rate of transplants reached above 90% with the best treatment.2. Simplied propagation technique: In the experiment of predigesting media, the important order of influence of culture medium compositions on differentiation is tap water > inositol > sucrose > mass element. By the treatments that cuttings were soaked in 20mg/L IBA for 20 min and 100mg/L for 15 min on rooting initiation was similar, the highest rooting rate was 77.3%. The rooting rate can reach 83.86% in plastic bag with sail medium.3. Microcutting propagation technique: transplanted tissue culture seedlings were used as cutting orchard. The branches were sheared and shaped, and the length of cuttings was 1cm. The best hormone treatment to cuttings was soaking 150mg/L of NAA+IBA for 5 min, the rooting rate was 93.75%. The best cutting time is May and June.4. Counting cost of seedlings: According to the routine method of tissue culture, the cost of tissue culture shoots was 0.27 yuan per seedling. However, the cost of tissue culture shoots is 0.19 yuan per seedling by predigesting procedure, which was declined by 29.6%.
