Acer maple‘Autumn Blaze'‘Autumn Blaze'(Acer×freemanii)belongs to aceraceae maple tree,native to the east coast of the United States,large deciduous trees,crown clean,leaf color fluorescent,autumn leaf color red marked,strong adaptability,is one of the excellent landscape greening tree species,the wood can also be high-grade wood floor.The red maple belongs to hybrids,the commonly used seed breeding easily mutates,asexual reproduction of vegetative organs can prevent mutation maintain its excellent characters,currently limited due to the introduction of seedlings,and grafting propagation and cutting propagation coefficient is low,cannot meet the demand of the seedling of large quantities of quantities,so seed micropropagation can effectively improve the reproductive rate.This research mainly carry out the following for this optimization study through the system optimization screening the following main results:The optimum culture of Acer culture was studied.It is easy to ignore pollution control in tissue culture test and production.The influence of environmental control on pollution was studied in the industrialized production of "Maple Maple" autumn flame tissue culture.According to the experiment and practice,we think that the comprehensive control method,i.e.ultraviolet lamp + ozone sterilization + alcohol spraying sterilization,is the best way to control pollution,and the pollution rate can be controlled at about 7%.t was found that the new bacteriostatic Yipeiling had obvious effect on adventitious buds,and it was not harmful to adventitious buds.It was suggested that Yipeiling should be widely used in production.Through the monthly change of the contamination rate in the culture room,it was found that the month with the highest contamination rate was July,August and September in the middle summer and autumn of a year.Therefore,in the management of the tissue culture room,we should do a good job in cooling,humidifying and cleaning up the germs in time.Study on Optimization Cultivation of Acer 'Autumn flame' multiplication culture.The following conclusions are drawn.1)In adventitious bud multiplication,MS improved medium was superior to other medium.The best acidity was 6.0 agar and 25 g sugar;2)The optimum concentration of CPPU was 0.6 mg/L,and the ratio of CPPU to IBA was 12:1,and the proliferation rate was the highest;3)By investigating the effect of subculture times on proliferation,it was found that the multiplication rate was lower in the previous subculture,then gradually increased,and gradually stabilized after 10 inoculations.4)By investigating the ratio of vitrification rate to increment rate,we conclude that the concentration of CPPU should be gradually lowered between 0.3mg/L and 0.5mg/L after 30 successive generations.Mainly through the cultivation of American Red Maple "Maple Maple" autumn flame transplant conditions optimization.By comparing the peat soil + perlite in the transplanting medium,it was found that the peat soil + perlite used for seedling cultivation had higher rooting rate and 21-day survival rate than the peat soil in Northeast China.Through the optimization of bottle seedling: When 1/2MS+NAA 0.05 mg/L+IBA 0.05 mg/L nutrient solution was used for direct irrigation,the rooting rate was increased to over 95% and the survival rate was over 85% in 21 days,Addition of sugar sources will increase matrix pollution.Through the optimization of micro environment humidity,it is found that microenvironment maintains the best rooting of 90% minimum stem segments.
