Two experiments were conducted to determine the optimum parameters of non-starch polysaccharides (NSP) enzymes(cellulase and pectinase)and proteinase (neutrase , alcalase)to hydrolyze the cottonseed meal (CSM) , and to evaluate the feeding value of the enzymatic hydrolyzed CSM by the cellulase and neutrase on the piglets.Exp.1Study on the Optimal Parameters of NSP Enzymes and Proteinaseto Hydrolyze CSM in VitroThe trial included three contents:1.By adopting one step enzymatic hydrolysis method in which the CSM was hydrolyzed by the NSP enzymes only, a serial trials were conducted to study the effect of cellulase or/and pectinase level and hydrolysis time on the production of reducting sugar.The results showed that: the production of reducting sugar were increased with the cellulase or/and pectinase level and hydrolysis time increased. The optimum enzyme level was 1400U cellulase per gram CSM or 1000U pectinase per gram CSM .The optical enzymatic hydrolysis time was 5 h. while there was synergetic effect between cellulase and pectinase, the optimum enzyme level was 800U cellulase per gram CSM and 600U pectinase per gram CSM and the optical enzymatic hydrolysis time was 5 h.2. By adopting two-step enzymatic hydrolysis methods in which the CSM was hydrolyzed first by NSP enzymes( cellulase was 800U per gram CSM, pectinase was 600U per gram CSM)for 5h , and then hydrolyzed by proteinases ( neutrase or alcalase ), a serial trials were conducted to study the effect of neutrase or/and alcalase level and hydrolysis time on the degree of hydrolysis (DH) and trichloroacetic acid-nitrogen solution index(TCA-NSI). The results showed that by hydrolyzed CSM first for 5h with the cellulase(800U/g CSM) andpectinase (600U/g CSM) , then following the hydrolyzed with neutrase or alcalase, the DH and TCA-NSI increased with the proteinase level and hydrolysis time increased. The optical neutrase and alcalase level were 500U /g CSM or 0. 032AU/g CSM respectively.The optimal enzymatic hydrolysis time was 5 h.3. By adopting two-step enzymatic hydrolysis methods in which the CSM was hydrolyzed first by cellulase or/and pectinase for 5h and then hydrolyzed by neutrase ( 500U/g CSM) for 5 h, A serial trials were conducted to study the effect of cellulase or/and pectinase on the DH and TCA-NSI. The results showed that the cellulase or/and pectinase will increase the the production of reducting sugar , DH and TCA-NSI. The optical cellulase or/and pectinase were 350U/g CSM or/and 250U/g CSM.Exp.2 Study on the Feeding Value of the Enzymatic HydrolyzedCSM on PigletsA single-factor design was employed to study the feeding value of the the enzymatic hydrolyzed CSM by cellulase(350U/g CSM) and neutrase(500U/g CSM) . 28 Durock X Large, 24+1 day old weaning piglets were randomly allotted to 4 treatments with 7 replicates per treatment and 1 pig per replicate. The treatments included soybean meal (SBM) basal diet, 12?%CSM diet, 15% CSM incubated under the same condition like enzymaticly hydrolyzed CSM but without enzymes, and 15% enzymatic hydrolyzed CSM. The trial was conducted for 21 day . Body weight gain, feed intake and diarrhea index were examined during the trial. The results indicated that 15% enzymatic hydrolyzed CSM did not improve the performance of weaning piglets, but reduced significantly the diarrhea of piglets. The higher free gossypol of enzymatic hydrolyzed CSM may be responsible for the poor effects of enzymatic hydrolyzed CSM.
