Wheat stripe rust, caused by Puccinia striiformis.f.sp.Tritici , is one of the most serious diseases to wheat production throughout the world. Genetic research to wheat stripe rust is a very important topic for the genetics and pathologists. Vilmorin23 is an important international differential host to wheat stripe rust and important international antigenic material, including resistance gene YrV23, Yr3. It takes on important theoretical and practical significance to molecular marker and gene location to genes in Vilmorin23. 125 F2 plants were inoculated by Puccinia strilformis.f.sp.Tritici 2E16.In all F2 plants, 101 resistance plants were found, and other 24 were susceptible. χ2 was 2.24, which is less than 3.84.Samely in all BC1 plants ,11 resistance plants were found, and other 6 were susceptible. χ2 was 1.77, which is also less than 3.84.So we can conclude that the resistance of NIL (nearly-isogenic-lines) Taichung29*6/ Vilmorin23 to 2E16 is controlled by one pair of dominant genes. Using the NIL Taichung29*6/ Vilmorin23 carrying the resistance gene YrV23 ,its recurrent parent and Vilmorin23 as materials, 55 SSR primers located on 2B chromosome were screened. Amplified DNA fragments were separated with 6% denaturing PAGE (polyacrylamide gel electrophoresis) and displayed by silver staining. Two polymorphic DNA fragments (Wmc356, Wms191) were found between NIL and recurrent parent, and the NILs amplified samely with Vilmorin23. Experiments were repeated and it proved the two pairs codominant polymorphic DNA fragments were reproducible. 150 F2 plants were used to analyze linkage and genetic distance between Wmc356 and YrV23. Wmc356 was confirmed to closely link to YrV23 with genetic distance 9.4cM.Samely Wms191 was linked to YrV23 gene with genetic distance 19.3cM. According to the SSR primers genetic map on 2B chromosome, and genetic distance of marker Wmc356, Wms191 to YrV23, YrV23 can be located precisely on 2B chromosome showed by diagram: Wmc356-YrV23-Wms191. Using the NIL Taichung29*6/ Vilmorin23 carrying the resistance gene Yr3 ,its recurrent parent and Vilmorin23 as materials, 69 SSR primers located on 1B chromosome were screened. We did not find codiminant polymorphic DNA fragments between NIL and its recurrent parent.
