Wheat is the most important food cereal plants in China and the drought becomes the main influential factor in the wheat production. Therefore, it is an urgent question to select drought-resistant wheat variety for excavating the wheat out-put potential in the drought and half arid area. Chemical mutation breeding is effective on inducing high frequency and wide scope mutations. If used in wheat breeding, it can bring all kinds of variations and rich wheat germplasms. In view of the fact that it is hard to find mutant wheat in the field and the environment has a strong effect on selecting drought resistant plants. So mutagenesis and in vitro culture were combined to breed drought resistant wheat in this research. The research studied on suitable concentration of PEG and EMS suitable concentration and got some mutations. Analysis of wheat drought-resistant mutant with SSR marker provide the theoretical fundamentals for chromosome location and cloning gene and breeding drought resistant wheat variety. The results are following:1 Drought resistance wheat (Hua84, Jin33) and drought susceptive variety (Wen6, Zhou 17) were choosed as materials, and their roots were observed in the same medium added PEG. The drought- sensitive wheat root appealed feeble growing tendency even no root under 80g/L PEG6000 water-stress. It is concluded that what the regenerated plantlets had root or not can be used to identify drought resistance under 80g/L PEG6000 water-stress.2 In this research, EMS was used to mutate the immature embryos, anther callus, . immature embryos callus of Wen6 and Zhoul7. About 50% relative initiating rate andrelative differentiation rate dose of the mutagens were used as the selective indexes of optimal mutagen concentrations and treatment time. The combination of treatments were as follows: Immature embryos treatment with EMS solution 0.80%+ 8h; Anther callus treatment with EMS solution 0.20%+ (2h~4h) ; Immature embryos callus treatment with EMS solution (0.20%~0.40%) + (2h~4h). The tolerance of Zhoul7 was higher on EMS than that of Wen6.3 The differentiation plants were planted in the rooting medium added 80g/L PEG, which were differentiated from EMS mutate materials, and 21 drought resistance plants...
