There is a lot of gene for wheat improvement in wild relatives Triticeae. Research works on it is meaningful to extend wheat gene resource and wheat breeding sustainable. Roegneria thoroldiana((Oliv.)Keng,2n=6x=42,StStPPYY)is the mode species in Sect.Paragropyron, Triticeae tribe of Gramineae. In 1990,Yan Ji,Yang Jun liang build up a new genus Kengyilia, all of Sect.Paragropyron mergered in Kengyilia. In china, thoroldiana grow in Qinghai and Tibet, possess drought and cold resistant gene and polyanthus,more tillering node, is the excellent gene reasource of wheat. For conservation and utilization of Roegneria thoroldiana, genetic diversity analyses utilize in situ hybridization, morphology and SSR in twelve Roegneria thoroldiana populations that collected from Qinghai and Tibet. The research results were as follows: 1) There is transposal of the chromosome in the thoroldiana. 2) There is a lot of genetic diversity of morphologic and SSR in thoroldiana. 3) Thoroldiana 77%genetic diversity within populations,the gene flow was estimated to be 0.83.That is the character of partial inbreeding plat. 4) Highly positive correlation was observed among SSR diversity analysis, altitude,location of collection and habitat,no correlation was observed among morphology diversity analysis, altitude,location of collection and habitat.But morphology clustering analysis is similarity to SSR clustering analysis,the populations which is clusted one group have the same altitude,location of collection and habitat. 5) The Tang gu la mountain of Tibetan plateau Thoroldiana populations have high genetic diversity. 6) Synthesis the SSR analysis result,in situ hybridization,morphologic to establish conservation strategy. we must priority consider the populations that grow in high altitude region,but can not ignore other populations. 7) The transposal material must priority protect and research.
