Genetic diverrity of wild Roegneria kamoji from Sichuan and Chongqing province as the central materals were determined using morphological markers, karyotype analysis and Gliadin markers.Main results obtained were as follows:1 Genetic diversity of 26 accessions of Roegneria kamoji Germplasm distributed in Sichuan and Chongqing province was studied in this paper.Twenty-four morphological traits were observed.The results showed that there was abundant genetic diversity among these accessions in morphological traits(H′=1.955).As for the genetic diversity index,it was mainly found within accessions(64.48%),while among accessions was 35.52 percent.To single accession,Fengdu Xincheng was the highest diversity index(H′=1.382).Twenty-four wild accessions can be clustered into four groups,the findings implied that a correlation among the tested wild resources, geographical and ecological environment.Principal component analysis showed that intemode length,upper glum length,the first lower palea spike length,spike length, leaf width,lemma length and florets per spikelet number were important traits in explaining the morphological diversity of the region.Additionally,effective conservation and utilization of Roegneria kamoji were discussed in this paper.2 Karyotypes of 12 accessions of Roegneria kamoji distributed in Sichuan and Chongqing province were studied in this paper.Among 12 materials,accession of Xichang Madao(RSC-273) has SAT,with karyotype formulae 2n=6x=40m(2SAT) +2sm.Accessions of RSC-001,RSC-164,RSC-200,RSC-241 and RSC-304 own to 2n=6x=40m+2sm,while RSC-065,RSC-146,RSC-198,RSC-252,RSC-325 and RSC-428 own to 2n=6x=42m.There were three karyotypes,accession of Chongqing Wanzhou belongs to 4A type,two accessions belongs to 1B type,which were the more evolutional ones.The rest were 4A type.The results suggest that there were chromosome differentiation to some extent in arm ratio,length ratio,asymmetry index and relative length in accessions.3 Acid polyacrylamide gel electrophoresis(A-PAGE) molecular markers were used to detect the genetic diversity among 42 wild accessions of Roegneria kamoji collected from four regions(Sichuan,Chongqing,Gansu and Japan).The followed results were obtained.(1) A total of 29 bands were detected in all accessions,89.66 percent were polymorphic bands.The Nei's genetic similarity coefficient of the tested accessions ranged from 0.448 to 0.966,and the average Nei's coefficient was 0.695. These results suggested that there was rich genetic diversity among the tested wild resources of Roegneria kamoji.(2) 42 wild accessions can be clustered into three groups.Moreover,the groups from the same origin frequently clustered into one group.The findings implied that a correlation among the tested wild resources, geographical and ecological environment.(3) Genetic differentiation between and within five eco-geographical groups of Roegneria kamoji was estimated by Shannon's diversity index,which showed that 83.69 percent genetic variance existed within group,and 16.31 percent genetic variance was among groups.(4) UPGMA cluster analysis based on Nei's unbiased measures of genetic identity was assayed for six eco-geographical groups of Roegneria kamoji,indicating that there was a correlation between genetic differentiation and eco-geographical habits among the groups.
