Roegneria shandongensis is part of Triticeae Dumort, widely distributed in esternChina, and most of them lived in a dry and barren soil, and plays an important role inecological and environmental protection. At the same time, Roegneria shandongensis isan excellent perennial grass with high economic value. Particularly important, theShandong Roegneria contains rich genetic resources of the high resistance to barleyyellow dwarf virus, is an important gene pool for wheat breeding.But now, relativelyweak in the Shandong Roegneria germplasm resources, there was no report aboutShandong Roegneria genetic diversity, genetic diversity studies will contribute to theprotection and rational use of germplasm resources. Materials collected from themainland and the islands of Shandong Roegneria in different populations, this study usedto study the genetic structure and differentiation,mainly used co-dominant molecularmarkers SSR and chloroplast genes trnL-rpL32to explore the mainland and the islandsof Roegneria grass genetic structure, genetic relationship and geographic diffusion paths,in order to reveal the Shandong Roegneria core of the plant germplasm resources, theformation and the impact of geographical isolation on genetic differentiation. Achievedthe following main results:1)Through research, we ultimately determine the PCR reaction was as follows:10×Buffer (Mg2+free)2.0μL,2.5mM MgCl22.0μL,2.5mM dNTP Mixture1.6μL,50ngDNA,2μmol/L primer3.0μL,1U Taq, glycerol1μL, betaine1μL, add water to cover to20μL.2) We have finally screened16polymorphic primers from183nuclear DNAprimers self-published in the barley and wheat, use of these primers to determine the Nabetween island and mainland populations of up to4.0000, minimum1.3125; Nemaximum2.7110, minimum1.3125; I maximum1.0246, minimum0.2166.3) Populationmaterials of Roegneria shandongensis show geographical distribution characteristics in the genetic. That is the populations which have adjacent location and similargeographical environment stay together. They can be divided into a ZhuCha islandpopulations, LingShan Island populations, because YangKou populations and TianHengisland populations have a close genetic relationship, can be attributed together;4)Although the Roegneria shandongensis populations inside the islands have a certaindegree of genetic differentiation from each other, more cases are manifestations of thereconsistency. A further research shows that, within these populations,they have a very lowdegree of genetic differentiation.5)Chloroplast genome phylogenetic analysis show thatRoegneria shandongensis plants have the same female parent donor, but geographicisolation and other factors have prevented gene flow between the different islands andthe mainland. This lead to the formation of specific genotypes.
