M3 generation and M4 generation of high flavone mutants from three tartarty buckwheat induced by 60Co-r rays was studied for genetic variation and analysis and determination of flavonoids, The purpose wae to ascertain the mechanism on improving of flavone content in buckwheat radioactivated and to provide scientific basis for selecting later generations and comprehensive utilization of the tartary buckwheat。 The results obtained were as follows:(1) The study was conducted to assess the genetic parameter on six agronomic characters of twenty-three mutants of tartarty buckwheat indicated that plant height, amount of branch and 1000 grain weight have high heritability I .e: 84.85%, 76.36%, 90.33% respectively; amount of kernel\weight of plant amount of have large genetic coefficient of variation .I .e: 20.13%, 16.06%, 19.33% respectively; Amount of kernel and amount of branch are improvered easily in the light of RGS.(2) Six agronomic characters of twenty-three mutants were studied by genetic correlation analysis and path analysis ,Result suggested that they have higher positive correlation from the relation between amount of branch, amount of kernel, 1000 grain weight and weight/plant. This showed thess characters have great genetic potentialities for increasing of weight/plant. Amount of branch and 1000 grain weight have higher herelidity (76.36and 90.33 respectively) and are used as indirect selecting character for weight/plant.3) By analysising correlation of flavone content and agronomic characters ,the results showed that the correlation coefficiency between plant height, main stem rode number, amount of branch , amount of kernel and flavone content is positive correlation, above all the correlation coefficiency between plant height and flavone content is 0. 5538>P0.01=0. 526.The correlation coefficiency between 1000 grain weight and flavone content is -0.7251, it is absolute value is bigger than P0.01=0.526, That is to say the higher plant height is and the less 1000- grain weight is, the higher flavone content is. Therefore, while selecting high flavone mutants. We should select populations of higher plant and lower 1000-grain weight consciously, there is more possible for the strains with the higher flavone content .(4) By analysising the zymograms of EST isozymes among the mutants of M4 generation, the results indicated that isoenzymes have some different, above all, mutants whose agronomic characters have greate change such as A2, B4, D3. Numbers, Colour and witdh of their zymograms have obvisely different. Isozymes is direct result of gene convey . its variation has correlation with hereditary character. Obvious difference in agronomic character of mutants and parents was caused by mutation.(5) Total content of flavonoids and component of flavonoids were detected by colorimetric method and HPLC respectively, The results showed total content of flavonoids in M3 and M4 is higher than parents and is not change obviously in M3 and M4. this indicated its heredity is stable after content of flavonoids is improved.Radioactivity cause hereditary variation, It is available to improve flavone content by radioactivivated tartary buckwheat.Four components of flavonoids were found in Seeds of B2 and Kp9920, and proportion of components is consistent, content of Rutin is the highest.
