Four different varieties of wheat (beiyi1,suyin10,lumai22,jinchun9) were treated by means of vernalization,devernalization,cold acclimation. Changes of soluble protein of them were analyzed by SDD-PAGE, and Analysis conclusion showed: a certain vernalization time for sprouting seed could increase the soluble protein content of wheat .After cold acclimation, 46kD protein content increased sharply,it had something to do with cold hardiness of seedlings. Wcs120 gene encode a 50kD protein specifically induced by cold acclimation. This protein similar with 46kD, the primer was designed according to the Wcs120 gene sequence. The primer region of Wcs120 gene was amplified and the regulation region of cold-induced gene was analysis. The results are as follows: The promoter sequence of Wcs120 are various in different wheat variety. The primer region sequence of cold-insensitive cultivar was coherent on the reported gene, but the primer region sequence of cold-sensitive cultivar was different from the reported gene. There were some lack of A-T while increase of G-C in the cold-sensitive cultivar. On 610 loci of amplify sequence of suyin10 and lumai22, there was a lack of ACG. These varieties may result in the different expression of the Wcs120 gene in various wheat cultivar.
