The optimum separating technology of polyphenols from the skin and core of the Ziziphus jujuba Mill.cv. Jinsixiaozao has been studied in the paper. Meanwhile, the antioxidant activity in vitro and physiological function of polyphenols from the two parts of the fruit to mice has also been studied. The main results are as follows:1. The optimum condition for extracting polyphenols from the skin and the core of the Ziziphus jujuba Mill.cv. Jinsixiaozao was determined to be degreased by petroleum ether at first, then kept extracted for 12 hours using ethyl acetate and distilled water (1:1) for twice, collected ethyl acetate phase, then vaporized to solid, redissolved the solid by ethanol to liquid and stored at -20℃. After that, Thin-Layer Chromatography has been done using ethyl acetate, ethanol and distilled water (1:1:0.3) as spread reagents. One band was gotten from the skin polyphenols, which R? was 0.8. but two bands were gotten from the core polyphenols, which R? were 0.62, 0.77 respectively.2. The extracted polyphenol specimen had been eluted by polyamide CC using 30mL of 30%, 50%, 75%, 90% ethanol at different periods. The different filtrates were collected and vaporized to minimum volume respectively, then redissolved by ethanol and stored at -20℃. The results showed that the polyphenols of the Ziziphus jujuba Mill.ev. Jinsixiaozao could get the maximum using 75% ethanol as elution, which the skin polyphenol was 80.64%, and the core polyphenol was 82.07%.3. We analyzed the polyphenols by HPLC after the millipore membrane, and the optimum condition was: we injected 10μL samples on 276nm at 30℃, which eluted at 1.0mL/min by pH=3.5 CH3OH:H2O = 25:75. Three components were gotten from the skin (tR1=2.52min, tR2=2.77min, tR3=4.19min);two components were gotten from the core (tR1=2.46min, tR2=4.20min). As the standards, tRcatechol=2.80min, tRphlorglucinol=2.49min. It indicated that catechol and phlorglucinol were probably in the skin, while phlorglucinol was in the core.4. The antioxidant activity of the polyphenols in vitro was determined by using BPCLL and UV. The results indicated that the polyphenols had inhibition to hydroxyl freeradical, inferior nitric acid rot, DPPH-, besides it has a better protective action to DNA damage caused by -OH.5. The anti-inflammatory, anti-cruor, endure-anoxic abilities, SOD activity, MDA content experiments had also been done by the polyphenols. It showed that the polyphenols from the skin and core of the Ziziphus jujuba Mill.cv. Jinsixiaozao had markedly effect on anti-inflammatory, anti-cruor, endure-anoxic, and could also enhance the SOD activity but decrease the content of MDA in vivo.
