The planting area of jujuba "JUNZAO" is more than 60%in Xinjiang,and it is the biggest planting scale of fruit trees.So far,the development and utilization of jujuba is focus on its fruits,the leaves after picking the fruit are downplayed,which causes a great waste of resources.The content of Flavonoids from jujuba "JUNZAO" was relatively higher in previous studies,it can be a major source of high-purity flavonoids in industrialization production,which can promote the exploitation of jujuba resources.Fingerprint of leaves from different varieties of jujuba was established in the study,leaves of jujuba "JUNZAO" were used as materials,ultrasonic-assisted extraction method was used for the extraction of total flavonoids and macroporous resin was applied to separate and purify flavonoids,the impact of various factors on the flavonoids isolation and purification was discussed to determine optimum process parameters.On this basis,the utilization of a variety of modern chromatographic methods was applied to obtain higher purity flavonoids date in the further separation and purification,and their structures were identified.The results showed as follows:1.Total flavonoids was extracted by ultrasonic-assisted by ultrasonic-assisted extraction method,the optimum conditions was determined by response surface methodology which based on single factor test:the concentration of ethanol was 60%,solid-liquid ratio was 1:30,extraction time was 50min,temperature was 55 ?,verification experiment was carried out in the condition,yield of total flavonoids was 15.87%actually.2.Macroporous resin was applied to separate and purify flavonoids in the further,various parameters were discussed.Results showed that AB-8 among six different macroporous resins was the best for the separation and purification of the flavonoids from the leaves of jujuba "JUNZAO",the pH of sample was 6 with80%ethanol as the eluted solvent and elution volume was 4BV.The elution ratio was 79.58%and the purity was 30.05%under the condition.The purification process can be simple and effective for enriching flavonoids from leaves of jujuba "JUNZAO".3.Fingerprint of 33 jujuba species was established in this study,cluster analysis showed that content of flavonoids from leaves of jujuba "JUNZAO" was higher than others.Nine common peaks in chromatograms were completely separated from other peaks,and the content of seven major peaks among them were determined.4.15 compounds were isolated and purified by AB-8 macroporous resin,silica gel column chromatography,gel column chromatography and Pre-HPLC.The structures were identified by physicochemical properties and spectroscopic methods as Quercetin(1),quercetin-3-O-p-D-glucoside(2),rutin(3),kaempferol(4),kaempferol-3-O-?-D-glucosidase(5),kaempferol-3-O-a-L-rhamnoside(6),stearic acid(7),lauric acid(8),scopoletin(9),glucose(10),sucrose(11),oleanolic acid(12),ursolic acid(13),gallic acid(14),uridine(15).
