Populus tomentosa is one of important forestation trees by means of asexual propagating in our coutry and all over the world. The clone reproductive capacity of inhomogeneity p. tomentosa is very different, what's more, the cyclophysis of the same tree in the same variety is very different,too. The tree's individual developing about maturation and rejuvenation including yang growth and majority of them, are attracting the people even more ,so it has been an important task.In the study we mostly analysed the difference on cyclophysis of different part of mature p. tomentosa from developing and principle biochemical process, using tissue culture, endogenous hormone's comparatively, and the technology of mRNA differential display .on the basis of predecessor studies ,we still thoroughly revealed the cyclophysis mechanism in wood plant individual,and explored the pass capacity of cyclophysis to offspring during asexual propagating,and resulting in nongenetive variation inner clone. The main results from this study are as fellows:(1) In this paper, bud from different part of p. tomentosa for 20 years cultivate in vitro. The result indicates that coefficient of bud formation and the growth of tissue culture durative increases. Leaf cultivate in vitro for 50 days, the result show that the number and the length of root rise from upside to grass roots of white poplar1 and 2 in turn. Shoots propagate in vitro for 40 days, the result show that the number and the length of root rise from upside to grass roots of white poplarl and 2 in turn. Mature degree minishs from upside to grass roots of grown poplar. Difference is markedness between grass roots and the other part .(2) The result of mensuration of endogenous hormone show the rate of IAA/ABA rise from upside to grass roots of grown poplar in turn;The content of IAA, ABA isn't orderliness in different part,GT is absent and content of ZT is steady in seedlings of tissue culture.(3) The rate of IAA/ABA rise from upside to grass roots of grown poplar in turn. The coefficient of bud formation and the growth of tissue culture, the number and the length of root of leaf cultivate in vitro is positive correlated with the rate of IAA/ABA, pertinence is 0.744. 0.9453, 0.9758, 0.9895, correlated rate is terribly markedness except 0.744.(4) The growth rate of tissue culture in the base part of a tree obviously is faster than thecrowns,it is correlated with the young developing period and strong genetic ability. Its inner mechanism possibly refer to genetic factor and the correlate matter .In this experiment ,through tissue culture ,differential display,we screened out one special cDNA strip on 453bp in the crowns ,it is probably relation to cyclophysis.Sequence analyzing ,the result has certain homology with NADH 1 and NADH V of camellia ,but the degree of homology is low,only 32 base pairs ,it is probably a kind of new gene ,and its encoded amino acid sequence and its function need more researth.
