Study on the Technique system of Tissue culture in Triploid Pop ul us tomentosa Ye Deliu (College of Forestry and Horticulture, Sichuan Agriculture University, Ya抋n, Sichuan, China, 625014) Abstract It is studied to find out the best composition of medium and propagation procedure with the explants of terminal buds, axillary抯 buds, leaves of Triploid Popzdus tomentosa The results were as follows: I )Culture conditions: temperature 25? 0C, 14 hours illumination every day, and about 2000 Lux illumination intensity. 2)The optimal explants were young leaves from Triploid Populus tomentosa in growing season. The structure of young leaf callus which was green-yellow was looser than old leaf callus, and this kind of callus was easy to differentiate to buds. 3)The best primary medium: II2MS+BAI.5mglL+NAAO.2mglL. After 80 days culture, buds were grown thickly. 4)Subculture medium: I /2MS+BAO. Smg[L+NAAO.O5mgi扡. Theoretically, the rate of propagation of leaves could reach about 5 for 24?6 days and 6.1 xl ~ for one year. 5)Using l/2MS in which any plant growth regulators was not added, stronger nurslings could be got. 6)Rooting medium: l/2MS+NAAO.O5mgIL+IBAO.5mgIL. The rooting rate could reach 98.8%. 7) For seeding rooted nurslings, the basic material consists of soil, sawdust and stool of chicken and duck, with plastic film cover, watering one time every day, and taking away the plastic film after 30 days, the rooted nurslings?alive rate could reach 100%. 29 8)Using leaves of rooted nurslings as explants, contaminative rate could be reduced, and propagation coefficient could be increased.
