Objective: By discussing the effect of Pollen-amylose of Maize (PAM) on the cumulation of Nitric Oxide, IL-6, ET-1 in the culture of Rat Intestinal Villus Microvascular endothelial cells (REC) when it enhance the immunity function, we expect to explain how the PAM work in immunoreaction as a helper- immunoreaction and probe into the basic work mode of immunity accommodate. Methods: Add PAM solution to the culture medium of rat intestinal villus microvascular endothelial cells, and then mensurate the cumulation of nitric oxide, of interleukin-6 and of endothelin-1 in the samples of the culture medium in different time. First, distilling PAM and sublimating it and determine it's content. Consult common mathods of plants amylose and upswing it. Grain alcohol was used directly as circumfluence, and then bring up with water and alcohol was used to sunk. The method of vitriol-hydroxybenzene was used to ensure the content of the PAM. The number was 78.3%. Second, cultivating rat's endothelial cells and sublimating and judge it. Consulting the normal blood sugar level, I designed the original chroma to be 100μg/ml. It maybe the one 600th or 1000th of the normal blood sugar level. The actual chroma was between 48μg/ml. When the cells developing about 80%, I digested and gained them and diluted it to 1×105/ml. Added 0.5ml to each whole of a new plant and then put it into 37℃. Add 20μl PAM solution into some whole by the designed scheme. And then sampling by the designed scheme. After 3000rpm×5min in centrifuge, the clear liquid in the sample was accepted and frozen. Then mensurated the content of nitric oxide, of interleukin-6 and of endothelin-1 in the samples of different teams. Result 1: The cumulation of nitric oxide in samples are more higher than the contrast and show degressive trend in 7 hours. This result show that PAM stimulating REC to excrete more NO suddenly and then reduce the output. It means that PAM can enhance the cumulationof Nitric Oxide from REC. Result 2: The cumulation of interleukin-6 in samples are more higher than the contrast and show the same rules with the contrast. Result 3: The cumulation of endothelin-1 in samples are more higher than the contrast after 48 hours and show elevatory trend. Conclusion: The PAM can enhance the immunity by influencing the secretion of the amount of nitric oxide, of interleukin-6 and endothelin-1 from REC. With the stimulation of PAM, the blood vessel put up dilation firstly, the clearance among the cells in the vas wall, it can enhance the inflammation;secondly the vas shrinking, it can control the leak from the blood vessel wall and lightenning the degree of the inflammation. The whole results shows that the cumulation of some cytokines(for example NO & ET-1) from REC can be changed by the simulating of PAM . When PAM enhance the immunity function, the movement of dilation and shrinkage concomitancy phenomenon maybe appears in the body. In other words, it shows that the mechanism of immunity enhance maybe PAM regulating the exudation function of REC firstly. So,we could suppose that the endothelial cells locate some key status in the whole immunity regulation system very possibly.
