The quantitative single radial immunodiffusion technique(RID) was applied to determine IgG levels in Alxa Bactrian camels colostrum as well as the extent of the calves'passive immunization by maternal antibodies. The results can not only make clear the pattern immunity obtained from maternal antibodies and but also help us to understand the factors of affecting the extent of the calves'passive immunity obtained from maternal antibodies. The milk IgG was acquired by precipitation with saturated ammonium sulphate and anion exchange resin such as DEAE-Sephacel, and it's purity and molecular weight were determined by SDS-PAGE.The specific rabbit anti-camel IgG antiserum was first prepared by injection of purified camel IgG into different sites of rabbits. Moreover, the RID was successfully established in this study: Milk samples from dams were taken immediately after delivery and 12,24,36,48,72h,5,7,14,30 and 90d later. The milk samples taken at the same stage of lactation were pooled. The IgG concentration in the colostrum of dams at the time of birth was 170.5mg/ml and it decreased to 78.7mg/ml at 24h post-partum. This decrease in IgG concentration of colostrum with the first 24h was significant. After 24h the IgG concentration continued to decrease markedly and was only 5.2 mg/ml after 90d. The average IgG content of colostrum was 55.94mg/ml during 90d after parturition. Blood samples from the newborn calves were taken immediately after birth but before the first colostrum intake, and additionally after 12,24,36,48,72h,5,7 and 14d.The IgG content of the pre-colostral calves serum was zero. The IgG concentration in the blood serum of calves increased significant with the first 12-24h.After the first 12-24h there was no further increase and the mean concentration then decreased slowly. However after 48-72h the IgG concentration increased again, until 120h, the IgG concentration became stable.Camel skim milk samples were heated at 65,75,85,100℃for 30min.Presence and identify of IgG were monitored using SDS-PAGE technique. Changes in activity of IgG was measured by RID method. The whole activity of IgG in reference values of cow milk was lost at 75℃/30min versus 68.9% in loss activity of camel milk IgG..
