Studies On Construction Of Regeneration System And Methods Of Transforming CBF Gene Of Three Kind Of Landscape Trees

Material in this study are Ligustrun lucidum Ait,Cinnamomum camphora Presl,Paulownia fortune. The result showed that several factors affected callus,shoot and root inducement of Ligustrun lucidum Ait,Cinnamomum camphora Presl,Paulownia fortune, including cultivars,plant growth regulators. A rapid and efficient regeneration system of three tree was established. The regeneration ability of Cinnamomum camphora Presland Paulownia fortune was better than Ligustrun lucidum Ait .A regeneration system from embryo and stem segment with axillary buds of Ligustrum lucidum was developed and the experiment is ready for the next stage of genetic transformation.The optimum media were obtained for the germination of embryo, callus formation, shoot regeneration and shoot rooting. On the media with 6-BA(0.5mg/l)+NAA(0.1 mg/l) +GA(0.1 mg/l), the of germination speed of embryo is highest , and the media with MS+6-BA_2.0+NAA_0.02 is fit the growing of stem segment .Aseptic explant is established from Northern China chilling tolerant Cinnamomum camphora Presl,and investigated that the optimal medium for lateral bud differentiation is MS medium su-pplemented with 6-BA 1. 0 mg/L and NAA0.1mg/L,and optimal mulitiplication culture medium is MS medium su-pplemented with 6-BA 5. 0 mg/L and IBA1.0mg/L,and some experiments were done on redifferentiation.The methods found the optimum medium for the bud induction of callus from leaves,stem and callus of leaves by perpendicular experiment, which consisted of MS medium with nine different concentrations of NAA,BA and IBA. They are MS+BA2㎎/L+NAA0.2㎎/L+IBA0.1㎎/L, MS+BA1㎎/L+IBA0.1㎎/L, MS+BA1㎎/L+ NAA0.1㎎/L+IBA0.05㎎/L.When we established a rapid and efficient regeneration system of three trees ,we use three ways to transform CBF gene,such as agrobacterium, Pollen-tube pathway, injected by the cold-resistant gene.
