Boletus sp. is an important part of forest ecosystem. Generally, most plant species form a kind of symbiosis with beneficial fungi, which is called mycorrhiza. Mycorrhiza is considered as such a fundamental part of plants that most species could not survive in nature without it .The research on interactions between EM fungi and trees can be summarized as follows : 1) Effects of EM fungi on biodiversity,stability and productivity of forest ; 2) Ecological-comprehensive effects of EM on forest system such as increasing the effective survival of trees ,improving soil structure and soil microbiology; 3) Growth response of trees to EM fungi. Most of them are precious medicinal and edible mushroom, which are of better trophic and unique medicinal value. Much attention has been paid to inducing and cultivating the fruit bodies of Boletus sp.at home and abroad. The key of cultivation is to batch producing of pure cultured mycelium, and the difficulty is that pure cultured mycelium grows very slowly. For these reasons, we isolated and identificated S. bovinus. we made detail investigations on cultivation of S. bovinus, production of exocellular enzyme and diversities of the microorganism on the surface and inside of S. bovinus. These provided clues to promote growth of pure cultured mycelium.This thesis includes two chapters that consist of four parts as follows.[1] Isolation and molecular identification of S. bovinusBy means of tissue isolation, pure cultured mycelium was obtained from fruit body of S. bovinus, which is a kind of ectomycorrhiza fungi and can not be cultured artificially.The traditional morphological methods for the identification of S. bovinus are problematical, so we used analysis of rDNA internal transcribed spacer (ITS) sequences to identify S.bovinus and establish teevolution tree for preliminary studying phylogenetic relationships of S. bovinus.[2] Study on the cultivation of S. bovinusThis part discussed the experiment about the selection by sifting of the nutrient medium of mother seed to Boletus sp.and liquid culture and stock culture of S. bovinus. The results showed that mother seed to Boletus sp. grows best on the compounding nutrient medium with glucose and peptone .During the liquid culture of S. bovinus, the mycelia of it grew faster. Tissues of S. bovinus were cultivated in 3 kinds of stock media. After two months, S. bovinus grows best on the compounding nutrient medium with wheat.
