Ganoderma lucidum is traditional medical fungus in china, and many studies were carried out on the activity and active component. Therefore, discoverage and reservation for the spawn resource was significant for its development and application. Immunologic tests, fementation codition and property of several exocellular enzymes were studied after the spawn isolation and identification.Spawn Isolation And Identification ITS of spawn isolated from Ganoderma lucidum of Mount zijin amplified and sequenced , and then the similarity (100%) beteewn the two ITS sequences (mycelium and fruiting body from G. lucidum ) indicated the success of the spawn isolation . The confidence is 99% in the phylogentic tree. The basidiospore from fruit body Ganoderma lucidum was observed through scaning electronical microscope, the typical character of basidiospore confirmed the verity of the species. In Vitro Immunologic Test on EPS From G. lucidum. The immunologic test showed that EPS can promote celiac macrophage swallowing neutral red, enhancing the celiac macrophage to kill tumor cell, boosting lymphopoiesis in thymus and sleepn. The result confirms that EPS can enhance immunity of the rat.Fermentation for Mycelium After corn powder and soy powder being confirmed the optimum carbon source and nitron source respetively through selecting test, an orthegonal test was desgined to achieve a optimum fermentation.Exocellular Enzymes Property A starch enzyme peak occurred in the fermentation, but the other two enzymes (CMC enzyme, xylan enzyme) did not show a obvious peak in the fermentation course. The laccase can show activity in a little wide pH spectrum. Wheat bran and peptone are optimum for production, and pH3.0-6.0 is also fit for laccase production.
