The characteristics of the vegetarian growth period, fructification growth period, heredity distance using molecular markers are studied for thirty-seven strains of Volvariella volvacea collected, to study the cultivation traits and genetic differences of the strains in depth. Molecular marker techniques of RAPD and SRAP are used to select parents, identification and screening for hybridization, to provide technological basis for the construction of the breeding programme for Volvariella volvacea.1. Comparison of the characteristics of vegetarian growth period.Strains growth, activity of extracellular enzyme, torlerance of temperature are studies. It is found that Strains SmV23, V23-2, AN30, V040728-2, V040729-1 of have extremely significant growth at 1% level; and Strains VG, VK, AN20a, V189,草茹 101 have significant activity of extracellular enzyme at 5% level; Strains AN15, AN20b, AN29, 草茹 102 have extremely significant tolerance of temperature at of 1% level.Appearing time, amount of nucleus, its forming and growth process of chlamydospore are studied, no differences among thirty -seven Volvariella volvacea is found.2. Comparing the characters of fruit-body growthYield, the appearances of the strains are compared, high yield strains are AN03, AN08, AN20a, AN35, V040728-2, V040729-1, VPN; there Strains maturing less than lweek areSmV23, AN20b, AN26b, VPN; Strains do not open easily: VK, V189,草茹 102. These are potential core strains for hybridizing of Volvariella volvacea.3. Researching the heredity distance through molecule markThirty seven Volvariella volvacea were analyzed through RAPD and SRAP, by the dark clustering analyze.core strains are found: AN03 , VK, V189, V040728-2, 草茹 102, VPN. Strains growth's situation is standard of fine strains. VPN×AN03, VK×V189, VK×VPN are choosed as the best groups for hybridizing of Volvariella volvacea..4. Choosing parent strains for hybridizationSix groups of hybridizing were researched by single-spore isolation, single hyphal choosing, appetency of hybridizing, single hyphal tip isolation and purification, pair parents and hybridizing. Six strains of hybridizing were choosing from 10 pairs of hybridizing and 100 single-spore strains.5. Identification and study of hybridized strainsOffspring, monad strains and parents are analyzed by RAPD and SRAP, results showed that Strain b is the offspring of AN29X AN34 , Strain d was offspring of AN29X AN30.
