In our study, two abnormal single spore isolates were hybridized and got a normal strain 2628. Single cell was isolated from tip of mycelium of 2628 and was cultivated. It looked like normal, so two abnormal single spore isolates are plasmosgamized. Hybrid strain 2628 was identified successfully with RAPD, SRAP. The fruit body of 2628 had better appearance and had higher production comparing with main cultivated strains. The production of strain 2628 was as twice times as main cultivated strain V23-2. Biology conversion rate of strain 2628 reached 33.4%. Fifty-five single spore isolates were gained from hybrid strain 2628. Phenotypes of 52 single spore isolates were polymorphic. Progenies of single spore isolates had genetic polymorphic with SRAP markers. Fifty-five single spore isolates were divided into three groups by cluster analysis. Hybrid strain 2628 was heterokaryon by identification of hybrid strain and genetic analysis of progenies of single spore isolates. Effective identification method of hybrid strain of Volvariella volvacea was created and constructed breeding model.Four SRAP genetic markers of two single spore were converted into SCAR marker on the basis of SRAP identification of two single spore 26 and 28 and hybrid strain 2628. Two markers especially belong to single spore 26 and others especially belong to single spore 28. Four markers can be used to identify hybrid strains and analyze genetic polymorphic of single spore isolates of hybrid strain.
