Chinese fir[Cunninghamia lanceolata(Lemb)Hook] is special timber in South China, charactered by fast-growing, good quality, high production and wide use. Chinese fir has an important place in the forestry of South China, therefore, it is actually significant to collect and identify its genetic resources and analyze its genetic diversity .For several years, different scholars have done a lot of research on the law of its genetic variation of different geographic provenance and made a lot of achievements. However, most of researches present are being carried out morphologically, physiologically and ecologically, seldom from the angle of molecular biology. As a result, the genetic variation law of different provenances and distributing setup of species groups haven't been found out so far. The combination of the damage of idioplasm resources in recent years, unclear source of seed provenance and clone in forestation and the confused management of forestation materials of Chinese fir leads to the result that it is hard to identify Chinese fir from outside shape or by the means of physiology cell marker.Aiming at the problems existing in Chinese fir breeding at present, this research made a choice of twenty-four geographic provenances, nine optimized clones and six parented-offspring and their hybrid offspring as researching materials. For the first time, the research analyzed genetic variations of different geographic provenances and carried out fingerprint identification of hybrid ISSR, using ISSR technique which is better than RAPD in polymorphism, repetition and the ability to identify genetic differences. The research also analyzed genetic backgrounds of different provenances, genetic structure and the relationship between provenances, and explored the feasibility of identifying fir clones and its hybrids with ISSR technique. Meanwhile, the research revealed the law of rich genetic, variations between geographic genetic resources of Chinese fir 'and the distributing setup of breed groups, which provided evidence for identification of hybrids, purity analysis and the management of optimized idioplasm resources. The main results are as follows.1. Faced the fact that it is difficulty to extract DNA from Chinese fir which containspigments, phenolic compound and quite a lot of sugar, the researcher used improved SDS method to extract DNA OF Chinese fir., which meets ISSR requirements in quantity and purity.
