Fujian has rich tea resources,as Fujian's important economic crops,has a long history of tea cultivation.At the same time,because of the long-term hybridization and tea cultivation,so that accumulated a large number of tea germplasm resources.In order to identify the germplasm resources of tea in Fujian and explore the suitable methods for the genetic diversity analysis.The research analysis 55 tested materials for genetic diversity of Fujian tea varieties.The IS SR,SCOT and iPBS molecular markers were used to analyze the genetic diversity of 55 tea materials,and respectively constructed 55 materials fingerprints with three molecular markers.The specific results as follows:(1)A total of 156 locis were detected in 55 samples by 11 ISSR primers,which 152 locis were polymorphic.The every primer was average amplified 14.2 bands and the percentage of polymorphic bands was 97.45%.The genetic similarity coefficient among the 55 materials were between 0.56 and 0.92,the average was 0.72,and constructed 55 tea varieties' fingerprints by the ISSR marker.The 55 tea materials about two groups,the Observed alleles number was 1.87,the effective alleles number was 1.54,the Nei gene diversity was 0.31,the Shannon's information Index was 0.46,the genetic differentiation was 0.05,and the gene flow was 9.99.When the genetic similarity coefficient was 0.68,55 tea resources are divided into three categories.(2)In this experiment,the SCOT marker was first time used to analyze the tea resources in Fujian,and established the SCOT-PCR system for Fujian tea resources.Screened 16 polymorphic primers from 38 SCOT primers,and used The SCOT marker constructed 55 tea cultivars's fingerprints.A total of 219 locis were detected in 55 samples by 16 primers,which 216 locis were polymorphic,the every primer was average amplified 13.8 bands and the percentage of polymorphic bands was 93.15%.The genetic similarity coefficient among the 55 materials was between 0.49 and 0.85,the average was 0.67.The SCOT marker analysis 55 test materials for a total of two groups's Observed number of alleles was 1.93,the effective number of alleles was 1.54,the Nei gene diversity was 0.32,the Shannon's information Index was 0.48,the genetic differentiation was 0.067,and the gene flow was 7.01.When the genetic similarity coefficient was 0.64,55 tea resources are divided into two categories.(3)In this experiment,the iPBS marker was first time used to analyze the tea resources,and established the iPBS-PCR system for tea resources.Screened 13 polymorphic primers from 48 iPBS primers,and used The iPBS marker constructed 55 tea cultivars's fingerprints(lines).A total of 216 locis were detected in 55 samples by 13 primers,the every primer was average amplified 16.61 bands and the percentage of polymorphic bands was 95.83%.The genetic similarity coefficient among the 55 materials was between 0.4444 and 0.8379,the average was 0.639.The iPBS marker analysis 55 test materials for a total of two groups's Observed number of alleles was 1.96,the effective number of alleles was 1.69,the Nei gene diversity was 0.39,the Shannon's information Index was 0.56,the genetic differentiation was 0.057,and the gene flow was 8.34.When the genetic similarity coefficient was 0.61,55 tea resources are divided into three categories.(4)The research show that the genetic diversity of the index and similarity coefficient by ISSR molecular markers detected were higher than the SCOT and iPBS markers,and the polymorphism of primer amplification was also the highest,it indicating that ISSR marker's polymorphism detection ability was higher than the other two markers.Using the primer combination method to construct 55 test materials of the fingerprints,iPBS markers build 55 tea fingerprints has the highest efficiency,Only 11 locis with 2 primers,While ISSR and SCOT were respectively used with four primers 16 loci and five primers 15 loci.The polymorphisms of ISSR,SCOT and iPBS markers were high and not much different,means three markers are suitable for the analysis of genetic diversity of tea resources.The ISSR+SCOT+iPBS marker combined with the three markers combines the advantages of a single marker,in the overall analysis of the test materials,the clustering distribution of tea resources is reasonable.In the context rich resources of the future,can combined several markers analyzed test materials.The diversity of the 55 test materials analyzed by the three different markers,reflecting the richness of Fujian tea resources,the DNA fingerprints of 55 test materials can be used for the identification of Fujian tea tree resources,and provide theoretical basis for tea resources crossbreeding.
