Study On Technique System Of Rapid Propagation Of Three Ribes Species In Vitro

'Ribes diacantha Pall', 'RibesKomarvvii Pojark' and 'Han feng' were used as matierals to be studied on the technique system of tissue culture and rapid propagation in this paper. Four stages were involved in the whole technique system, including establishment of clone system, multiplication of shoots, induction of callus and rooting . The clone systems in vitro of the three types of Ribes were established for the first time, which would be the basic of further research by means of biotechnology in the future. The results are shown as below:1 Shoot of three Ribes were selected for water culture. Bud-stem or tender tip of Ribes were used as explant. It revealed that soaking explant by HgCl₂(0.1%) and two tip of tween for 8 minutes could sterilize explant well with polluting rate of 55%;Or soaking explant by Gentamycin Sulfate 1 (100mg/L) for 8min,and sterilizing explant by HgCl₂(0. l%) for 8 min could also sterilize explant well with polluting rate of 10%. The initial culture medium of three Ribes was MS supplemented with BA1.0mg/L and IBA0.1mg/L.2 Using the three species of Ribes,the experiment of bud multiplication showed that:The best sub-culture medium for Ribes diacantha Pall in vitro was MS medium containing BA 1.0mg/L and IBA0.05-0.2mg/L. The best sub-culture medium for Ribes Komarvvii Pojark and 'Han feng' in vitro was MS medium containing BA1.0mg/L andGA₃ 1.0-2.0mg/L. The period of subculture was suitable for 25-30 days, and the density of inoculation was 3-5/bottle.3 AgNO₃ can prevent Ribes shoots from browning were investigated in this experiment. The result was showed that 0.5-5.0 mg/L AgNO₃ can prevent shoots browning effectively with browning rate of below 20%.4 For three Ribes, common sugar could be used as carbon source in instead of analyzable sucrose of MS medium .5 Leaves and leafstalk of three Ribes could be used to induce callus respectively. For Ribes Komarwii Pojark and 'Han feng', best result of callus induction with leaves was obtained on MS supplemented with 2,4-D 0.8-1.0 mg/L,The best media for the callus multiplication of Ribes Komarwii Pojark were: MS+2,4-D1.0mg/L, BA1.0mg/L+2,4-D0.5mg/L, MS+BA0.5mg/L+2,4-D1.0mg/L+CH500mg/L or BA0.5mg/L +2,4-D1.0mg/L.For Ribes diacantha Pall, the best result induction of leaf was obtained on MS containing BA 0.5mg/L+2,4-D 1.0mg/L and BA 2.0mg/L+2,4-D 2.5mg/L.The best result induction of Ribes...
