In order to find out new techniques in tissue culture of Siraitia grosvenorii (Swingle) C. Jeffery, the Qingpiguo variety was used as material in the comparative studies of sterilizing effects on explants from different places, effects of different light condition on direct differentiating abilities of explants ,direct differentiating abilities of different explants , as well as the physical effects of different CTK in tissue culture of Siraitia grosvenorii. The results were shown as follows:(1) Better sterilizing effects could be obtained by using explants from near net house than from far fields. By using plant chemical regulation technique, our research team has successfully made Siraitia grosvenorii blossom and fruit in Nanning city of Guangxi . As a result, abundant germ-free plantlets can be obtained during a short time in Nanning in future.( 2 ) Cultured on the medium MS+BA2.0mg/L+IAA0.1mg/L, the middle part of leaves could directly differentiate regenerated plantlets under warm-color light with high color rendering (Ra≥80, the luminous flux was 3350 Lm, the luminous intensity was between 1200-1500Lx) , but couldn't while cultured under white light (the luminous flux was 2500 Lm, the luminous intensity was between 1550-2000 Lx).(3 ) Under warm-color light with high color rendering, compared to patioles and stems without buds , leaves , especially the base parts of leaves had strongest ability in direct differentiating regenerated plantlets.
