Cyclocarya paliurus is an endemic rare plants in China. In this study, the young embryos ofCyclocarya paliurus were chosen as explants and preliminary studies was performed on asexualreproduction of C. paliurus using tissue culture technology. The primary results are as follows:1. Young embryos of C. paliurus as explants were relatively easier for disinfecting. Thetype and concentration of the disinfectant has no significant influence on the germination andcallus induction, of the embryos, and disinfection with70%alchol for two minutes was the mostsimple way.2. In vitro culture, it was found that the middle stage of the embryo cotyledons wassuitable for embryo culture during using five different development stages of young embryos asculture materials, and germination and seedling rate reached100%and76.67%respectively.WPM is the best medium in five basic medium, where the highest rate of seedlings was hit to35%. No addition or adding too much GA3had an adverse effect on germination and seedlingdevelopment. Adding5.0mg/L GA3into the culture medium could obtained shighest seedlingsurvivals, reaching36.67%. Addition of the sucrose, the glucose or the mashed could improvethe germination rate of embryo and seedling survival. However, adding20g/L sucrose can get thebest germination rate of embryo and seedling survival, reaching more than80%and45%respectively. Addition of the TDZ0.1mg/L had the optimal induction effects on Clumpy buds, inwhich the multiplication coefficient reached6.71.3. During other factors remaining unchanged, if AGAR content of the medium increasedfrom0.60%to0.65%, the content of sugar increased from3%to4%, and the concentration ofthe NH4+reduced by half, the average of vitrification seedling dereased. However, when Ca2+content reduced half, the vitrification seedling increased.4. Rooting of tube seedlings was inducted by the method of step dipping. Dipping tubeseedlings in80mg/LIBA for75min, and then moving them to uncontain any hormone in aquarter of the DKW for culture, the rooting rate could reach20.83%.5. Using young embryos of Cyclocarya paliurus as explant to induce callus, the optimalinduced stage of young embryo was the middle of cotyledons and medium was the modifiedDKW, where the highest inductivity was88.89%. The combination of0.2mg/L2,4-D and2.0mg/L KT in plant growth regulator significantly improved the inductivity, reaching80.00%. MSwas the optimal prolifertation medium, but the increment and increasing rate of callus was thehighest during adding20g/L sugar in medium.1.0mg/L2,4-D+0.5mg/L KT was the bestcombination of plant growth regulator, whose average increment and increasing rate of calluswas9.27g per bottle and1363.24%respectively.
