The phylogeny of genus Epimedium involving in 12 species 26 populations in these study were studied on cytology, POD isozyme, interspecific crossability, officinal composition determination and their extracting technics. The results were as follows:The karyotype analysis showed that karyotype of the barrenwort species are similar to each other, with one pairs of m satellite chromosomes and symmetric karyotype. Among them, the karyotype formulas of E. leptorrhizum is 6m+4sm+4m (SAT), the rest all 4m+6sm+2m(SAT). The C-bands of 6 species native to Guizhou, China are different from each other, which were used for identification of barrenwort species sections.The zymograms of POD isozyme showed that 11 species have totally 16 bands and are different from each other.The crosses among 7 Epimedium species native to Guizhou, China showed that there are high crossability among these species, with the range 22.28-90.51%, indicating that they are close to each other. According to the crossability, they can be clssified as two groups: Group I, including E. myrianthum and Group II the rest. The flavones and icariin contents of wild E. acuminatum,E. wushanese and E. myrianthum are up to CCP (2005)'s officinal standard and those of E. acuminatum cultuvated in Guizhou Arboretum are not. There are significant differences of total flavones and icariin contents among the different populations of E. acuminatum.The effects of 2,4-D, 6-BA, NAA and IBA on leaf callus inducement of E. acuminatum were studied by means of L9(34) orthogonal design. The result showed that IBA and 2,4-D have the significant effect to induce barrenwort leaf callus. The order of the effect of the four hormones was IBA>2,4-D>6-BA>NAA. The good medium for barrenwort leaf callus inducement is B5 + 2,4-D 2mg/L + IBA 1mg/L.
