Gynura divaricata?L.?DC.Is a plant of the genus Asteraceae?Gynura Cass.?,Which is a common edible plant of medicine and medicine.Because of its strong pharmacological effect,it is widely used in the treatment of high-risk diseases such as hypertension and diabetes.However,due to the influence of factors such as ecological environment,cultivated area,and low natural reproduction rate,its extensive planting and comprehensive utilization are restricted.In this study,the stem segment was used as the initial material to establish an in vitro rapid propagation system;the tissue culture seedling leaves were used as explants,and the basic culture medium,plant hormone types and concentrations were studied by orthogonal analysis,single factor variables The influence of other factors on the tissue culture of Gynura divaricata?L.?DC.and establish the regeneration system of Gynura divaricata?L.?DC.And on this basis,the karyotype analysis process of the Gynura divaricata?L.?DC.chromosome was optimized.The results are as follows:1. Stem section propagation in vitro:The stem section of Panax notoginseng germinated on MS medium supplemented with 6-BA0.5mg·L^-1and NAA0.3mg·L^-1after 75%alcohol 30s+0.1%Hg Cl₂6min Buds work best.2. Establishment of callus regeneration system:the recovery rate of leaves in MS+6-BA2.0mg·L^-1+NAA0.2mg·L^-1medium reached 96.3%;MS+6-BA3.0mg·L^-1+NAA0.1mg·L^-1medium can obtain a higher adventitious bud proliferation rate;rooting rate reaches 93%in 1/2MS+NAA0.2mg·L^-1medium.Transplanted into a mixed soil matrix of sand and autoclaved soil grows well,with a survival rate of97.3%.3. Chromosome karyotype analysis:The root tips of Panax notoginseng were pretreated with 0.2%colchicine at 18?for 3h.After fixed for 24 hours with Carnot fixed solution,they were placed in 1mol·L^-1HCL at 60±0.5?The effect of dissociation under water bath for 9min is the best.The number of chromosomes in the white-backed panax notoginseng is 2n=18,the karyotype formula is 2n=2x=18=16m+2sm,and the asymmetry coefficient is 58.7%.belonging to the more primitive type 1A.
