Wheat–Th. intermedium alien disomic addition line II-1-1, resistant to powdery mildew was developed by Yannong15×Thinopyrum intermedium. In the progenies of disomic addition line II-1-1 crossing with the addition line Nonglin26 conferring 3C gametecidal chromosome of Ae.Triancialis and the ph1 gene mutation of common wheat Chinese spring, cytogenetics and molecular cytogenetic methods combined with resistance identification were used to identify wheat-Thinopyrum intermedium germplasms which had chromosome number of 2n=42 and resistant to powdery mildew. Results were obtained as below:1. Eight germplasms 03008, 03012, 03017, 03084, 04060, 04119-2, 04143 and 04146, resistant to powdery mildew were derived from 158 lines. Among them, 03008, 03017 and 03084 had chromosome number of 2n=44 and they were immune to powdery mildew. 03012, 04060, 04119-2, 04143 and 04146 had chromosome number of 2n=42 and they were highly resistant to powdery mildew.2. Chromosome configuration at PMC MI of 03012, 04060, 04119-2, 04143 and 04146 were observed and they all formed about 21II at PMC MI, and only seldom univalent appeared in few cells, which showed they were stable in cytology.3. Chromosome configuration of hybrid F1 between 03012 and Yannong15 at PMC MI was 2n=2.44I+19.78II, about two univalents appeared in every cell, which indicated that 03012 was a probable wheat- Th.intermedium substitution line.
