Wheat is one of the important food crops in the world, which plays an important role in the history of agricultural development. In recent years, due to the cultivation of wheat for directional selection and focus on the use of certain parent for breeding of wheat, the wheat quality instable, yields remain stagnant, ability to adapt to the external environment, the genetic basis of decline increasingly poor condition. To adapt to the modern agriculture of high quality and high yield of wheat genetic resources demand, only using their own genetic resources has been unable to meet the requirements of modern breeding in wheat production, so development and application of new genetic resources in wheat production has been imminent. Through transfer the good genes of wheat related species into wheat by distant hybridization, has become one of the main ways of solving narrow genetic basis of Wheat. Thinopyrum intermedium, one of species of third gene pool of wheat, contains rich genetic variations, played a important role in improvement of wheat for disease resistance, yield, quality and so on.The stable wheat line 08-738 derived from the progenies of chuanmai107 and Thinopyrum intermedium, and had shorter plant height and more spikelets per spike than its recurrent wheat parent. In this study, in order to identify the chromosome constitution of the line 08-738, analyses on the agronomic traits, the chromosome constitution of PMC MI, C-banding, genomic in situ hybridization (GISH), Fluorescence in situ hybridization(FISH) and SSR marker on wheat line 08-738 were carried out. The results are as follows:1. The average height of 08-738 is 76.5cm, average spikelet number per panicle was 23, and compared to the parent chuanmai 107 (86.8cm), it is 10cm lower, has more spike and it also has much more dense spikelet of the wheat top2. Observing the pollen mother cells meiosis metaphase I (PMC MI) chromosome configuration showed that chromosome pairing is 2n=42=21 II at PMC MI, and there is not find other chromosome pairing configuration, is suggested that 08-738 is a cytologically stable line.3. No apparent change of chromosome constitution was observed by Giemsa-C banding. The sequential GISH and FISH analysis indicated that the chromosome constitution of line 08-738 consisted of 20 pairs of wheat chromosomes and one pair of small segmental translocation chromosomes of wheat and Th. intermedium, and the translocation segment was closed to telomere of wheat 3DS and might be derived from the Js genome.4. The SSR marker analysis showed that SSR primer xcfd141 located on 3DS 6-0.55-1.00 could amplify one specific band between line 08-738 and Th.intermedium. Thus, primer xcfd141 could be used as the marker to select and identify this small translocation segment in wheat background.
