Lactarius deliciosus is an edible mycorrhizal fungus. It was chosen to be studied on tissue isolation, solid pure culture and rDNAITS sequence molecular identification. The main results of the research paper are as follows:1. The tissue isolation of Lactarius deliciosus is easy contamination so its success rate is low. Some outlines about the isolation are introduced. ①Basidiocap should be healthy and normal shape. Stipe should be thickset and solid. ②The tissue isolation of Lactarius deliciosus is operated as soon as possible after collecting Lactarius deliciosus. ③Skilled manipulation.2. The optimal conditions for the mycelial growth of Lactarius deliciosus were investigated. PDA or PDA synthetic medium are the best medium, the optimal temperature and pH are 26°C and 5.2 respectively. Fructose and NH4NO3 are the best utilized carbon nutrition and nitrogen nutrition respectively.3. The rDNAITS sequence of isolate of Lactarius deliciosus was sequenced. Molecular identification was accomplished using phylogenetic tree analysis of 22 Lactarius nrDNAITS nucleotide sequence including the 1 new and 21 GenBank sequence.
